With the rapid development of social economy,science and culture in our country,people's living standards have been continuously improved,and their health concepts have also undergone tremendous changes.There is a higher level of demand for quality of life,so that the health care products have become a sunrise industry with great development potential along with the increase of social demand.Health care products that contain functional ingredients extracted from natural products are increasingly popular with consumers.Moringa oleifera leaves have become a hot topic in current research because of its rich nutrition and medicinal value.Relevant reports indicate that there are many related foods that are marketed with Moringa oleifera leaves,such as tea,can,enzyme and noodles.However,there is no domestic health care product marketed with Moringa oleifera leaves as a raw material for sales.This study is based on the profound theoretical foundation of traditional Chinese medicine and many years of clinical experience,aiming at constipation and insomnia in sub-healthy people.It is mainly composed of Moringa oleifera leaves and supplemented by traditional Chinese medicine.It has designed and developed Compound Purgative and Compound Sleep Improvement based on Moringa oleifera leaves with innovative and strong market competitiveness.Objective and Methods1.Function study of Compound Purgative Moringa oleifera:Loperamide hydrochloride was administered at a dose of 6.67 mg/kg per day by gastric infusion for continuous 2 weeks so that constipation model was established.The effects of different groups of Compound Purgative Moringa oleifera on body weight,fecal number and water content within 24 hours,intestinal propulsive rate,contents of motilin,substance P,vasoactive intestinal peptide and somatostatin were observed by intragastric administration to explore the mechanism in the treatment of constipation at the level of gastrointestinal hormones,and to determine the best compatibility of Compound Purgative Moringa oleifera.In order to reduce the dosage,the best compatibility of Compound Purgative Moringa oleifera was refined by ethanol precipitation method.To ensure the its effectiveness,the constipation mice model was duplicated.Taking the time of defecation,the number of feces and the weight of dried feces within 4 hours as indicators,the effects of refined treatment were studied.2.Extraction and preparation technology of Compound Purgative Moringa oleifera:The extraction technology of Compound purgative Moringa oleifera was optimized by central composite design-response surface method.The content of isoquercetin and astragalin,the yield of dry extract and overall desirability(OD)were used as the evaluation indexes,the factors such as water addition,extraction times and time for extraction were investigated to determine the optimal water extraction technology.Because the yield rate was still high after refining,granules were chosen for the compound formulation.Taking the formability,fluidity,dissolubility and hygroscopicity of granules as evaluation indexes,the influence of different excipients and dosage on granules was investigated by comprehensive scoring method to screen out the optimum preparation process.The quality research method was preliminarily established by the content of the indicator components and total flavonoids,and TLC identification of the granules.3.Function study of Compound Sleep Improvement Moringa oleifera:Sleeping experiment was conducted in mice.Sleeping duration induced by pentobarbital sodium and sleeping delitescence induced by barbiturate sodium were recorded in each group.The effects of different groups of Compound Sleep Improvement Moringa oleifera on sleep improvement were observed by intragastric administration,and the best compatibility was to be determined.In order to reduce the dosage,ethanol precipitation method was used to refine the optimum compatibility of Compound Sleep Improvement Moringa oleifera.To ensure the its effectiveness,sleeping duration and delitescence were still employed as indicators to investigate whether refined treatment would have an impact on sleep improvement.4.Extraction and preparation technology of Compound Sleep Improvement Moringa oleifera:The extraction technology of Compound Sleep best compatibility ratio of Moringa oleifera leaves:Astragali Radix:Atractylodis macrocephalae Rhizoma was 15:4:6.Refining treatment had no significant effect on the purgative effect of the compound.2.Optimal extraction technology of Compound Purgative Moringa oleifera was as follows:3 times extracted with 10-fold the amount of water for 1 h each time.The granule with lactose as excipients was superior to the rest of excipients.The optimized conditions were as follows:the amount of dry extract powder:lactose as 1:1,wetting agent of 80%ethanol,and the flavoring agent was 0.3%aspartame and 0.2%orange flavor.The formability,liquidity,dissolubility and anti-hygroscopicity were good,and the particle passing rate was high.The contents of isoquercitrin,astragalin and total flavonoids were 0.58%,0.32%,2.67%,respectively.3.Compound Sleep Improvement Moringa oleifera could prolong the sleeping duration of mice induced by pentobarbital sodium(P<0.05),shorten the sleeping delitescence of mice induced by barbital sodium(P<0.05),and had a certain effect of improving sleep.The best compatibility ratio of Moringa oleifera leaves:Ziziphi spinosae Semen:Salviae miltiorrhizae Radix et Rhizoma:Polygalae Radix:Schisandae chinensis Fructus was 15:8:4:3:3.Refining treatment had no significant effect on the sleep improvement effect of the compound.4.Optimal extraction technology of Compound Sleep Improvement Moringa oleifera was as follows:3 times extracted with 12-fold the amount of water for 1 h each time.The granule with lactose as excipients was superior to the rest of excipients.The optimized conditions were as follows:the amount of dry extract powder:lactose as 1:1,wetting agent of 80%ethanol,and the flavoring agent was 0.4%aspartame and 0.2%orange flavor.The formability,liquidity,dissolubility and anti-hygroscopicity were good,and the particle passing rate was high.The contents of isoquercitrin,astragalin,salvianolic acid B and total flavonoids were 0.43%,0.25%,0.25%and 2.17%,respectively.Conclusion and Significance:Compound Purgative Moringa oleifera had a good laxative effect,and it is associated with adjustment of some gastrointestinal hormones.Compound Sleep Improvement Moringa oleifera had a certain sleep improvement effect.Central composite design and response surface method has the advantages of fewer experiment times,higher precision and fitting coefficient and better predictability.The extraction technology screened by this method is reasonable and feasible,and the quality of the preparation is stable and controllable.This study provides theoretical support for its clinical application and scientific basis for its development as a health care product,enriches the research content of health care products of Moringa oleifera,and is of great significance for making full use of natural plant resources.
