objective:To investigate the effect of musk and frankincense on the expression of prostate Claudins gene in C57BL/6,our group was carried on the further experimental research with it.Method:72 C57 mice(8-12 weeks)were randomly divided into 4 groups:the musk,frankincense group,musk combine frankincense group and saline control group,each group have 18 rats,each mouse was injected with 0.5mL in the corresponding group,and then sacrificed at 9h,24 h,48.The prostate tissues were dissected and collected for real-time fluorescence quantitative detection,and detected the expression levels of Claudin-1,-2,-3,-4,-5,-7,-8,and-10.Result:At 9h,musk could down regulate the protein expression of Claudin-1,Claudin-3,Claudin-5,and Claudin-8,the difference was statistically significant(p<0.01),frankincense can down regulate the gene expression of Claudin-1,Claudin-2,Claudin-3,Claudin-5,and Claudin-8(p<0.01),Meanwhile,the expression of Claudin-10 protein could be down regulated(p<0.05),musk combine frankincense group can down regulate all the Claudins(p<0.01)but Claudin-7,and the down-regulation of Claudin-5(p<0.05)and Claudin-8(p<0.01)genes was significantly different from musk group and frankincense group;At 24 h,musk combine frankincense group can increase Claudin-2,Claudin-3,Claudin-5 gene expression,the difference was significantly different(p<0.01),at the same time,the gene expression of Claudin-8 could be down regulated(p<0.05);At 48 h,musk could significantly up regulate the expression of Claudin-2,Claudin-3 and Claudin-8,and the difference was statistically significant(p<0.01),Frankincense can up regulate the gene expression of Claudin-5(p<0.01),musk combine frankincense group can increase the expression of Claudin-5 gene(p<0.05).Conclusion:The expression and regulation of Claudins protein in prostate of normal mice were determined by musk and frankincense,but no significant effect on Claudin-7 expression,in addition,musk,frankincense may also exist in the role of catalyst,the expression of Claudin-4 in prostate tissue is very low,and its role needs further study.
