Objective:To explore the effect of corkkonen on the improvement of atherosclerosis in Apo E-/-mice,and the effect of the expression of gsk-3 beta and wnt7a protein.Methods:68 Apo E-/-mice,according to the mice's weight and gender,were divided into Apo E-/-blank control groups of 8,and 60 in the AS model group.After the success of the model,the method was divided into the model group,low,middle,high dose nursing group,atorvastatin calcium tablet group,WNT inhibitor group.The wild mouse 8 was a control group for the general blank.The two control groups were given normal irradiated feed,and the rest were given high fat feed?21per cent fat and 0.15 per cent cholesterol?for 13 weeks.After the success of the model,low,middle and high dosage mindarmor kang group respectively for the 21st.6 g/?kg,d?,43.2 g/?kg,d?and 86.4 g/kg?d?.The atorvastatin group of 3 mg/kg.?d?,intraperitoneal injection of Wnt inhibitors,2%DMSO+30%PEG300+5%Tween80,200 mu g/ml,5 mg/kg,once every two days injection,the rest of the related drugs are batter made suspension liquid,soluble in distilled water to fill the stomach volume of 0.5 ml/day,1 time/day,for four weeks.After 17 weeks to be put to death in mice,mice thoracic aorta plaque,HE staining and Masson staining and immunohistochemical,respectively to observe the morphological changes and the expression of GSK-3 beta and wnt7a quantity.Results:To observe the morphological changes of the thoracic aorta with the naked eye:the two control groups of the aorta have no wrinkle,no lipid deposits,smooth muscle,and no foam cells.Model group aortic intima thickened obviously,smooth muscle direction change,is a large number of foam cells,HE dyeing is a large number of orange patches,Masson staining of a large number of green collagen fiber,immunohistochemical numerous brown granules.In the statin group,the medial dose of the protective group of the aorta was thickened by two blank control groups,the smooth muscle was incomplete and the foam cells were visible.The high dosages were thickened by two blank groups,and a few froth cells were visible,and the smooth muscle was incomplete.The lining of the inhibitor was thickened less than the other groups,so there were a few froth cells,and the smooth muscle was changed.There was no difference between the low dose group and the model group.Immunohistochemical method detects the effect of kantang on the expression of gsk-3 beta and wnt7a.Compared with the control group,the other groups of gsk-3beta and wnt7a were significantly reduced?p<0.01?.Compared with the model group,the gray value of the group,statins and WNT inhibitors were significantly higher in the group than in the model group?p<0.01?.The WNT inhibitor group was higher in grey than the high-dose care group?p<0.05?;There was no difference between the low dose care group and the model group;There was no significant difference between the medium dose care group and the statins.Conclusion:1.The heart can inhibit the formation of atherosclerosis plaques.2.The protective heart can significantly reduce the content of wnt7a and GSK's beta protein,which is resistant to atheroma The effect of this is probably related to this.
