Objective: To explore the effects of Sishenwan on gastrointestinal motility,Ghrelin and its receptor(GHSR)protein and m RNA expression with hypothalamus,duodenum in Asdthenic Splenonephro-yang diarrhea model rats.Methods:72 SPF SD rats were randomly divided into normal group,mode group,Dicetel group,Sishenwan high,medium and low dose group,with 12 rats in each.Except the normal group,other groups were treated with intragastric administration of adenine and Senna water decoction to replicate model rats with diarrhea of Asdthenic splenonephro-yang.After continuous administration of 14 d,the general state of rats was observed.diarrhea index,gastric motility and transit function of intestinal movementwere examined.ELISA method was used to detect the content of Ghrelin in duodenal tissue,Blot Western and RT-PCR to detect Ghrelin,GHSR protein and m RNA expression in the gastric and duodenum.Results:(1)Compared with the model group,the diarrhea index in Sishenwan high,middle,low dose and Dicetel group were gnificantly decreased(P < 0.05),and Sishenwan high dose group is particularly evident(P<0.01).(2)Compared with the normal group,in model group the Gastric residual rate decresed,the Intestinal propulsive rate increased,and the difference was statistically significant(P < 0.01).Compared with the model group,Sishen wanl each dose group and Dicetel group can inhibit rat gastric emptying and small intestinal propulsion in different degrees(P < 0.05 or P < 0.01),and Sishen wan high dose group was significantly better than the other groups(P <0.05),while shishenwan,low dose group and the Dicetel group had no significant difference(P > 0.05).(3)Compared with the normal group,the Ghrelin contents in model group significantly increased(P<0.01),The content of Ghrelin in each group were lower than those in model group(P < 0.05 or P < 0.01);Sishen wan high dose group was significantly better than in the low dose group(P < 0.05),there was significant difference between Sishen wan low dose group and Dicetel group(P < 0.05),while high,Sishen wan medium dose group and Dicetel group hadno significant difference(P > 0.05).(4)compared with the normal group,model group,the expression of Ghrelin in gastric antrum,duodenum and GHSR protein were significantly higher than normal group(P < 0.01);compared with the model group,Sishen wan high,medium and low dose group and Dicetel group expression can down regulate the gastric antrum,Ghrelin and GHSR protein in the duodenum(P < 0.05 or P< 0.01),and Sishen wan high dose group of Ghrelin and GHSR in the gastric antrum and duodenum,the expression of Ghrelin protein was significantly better than the other groups(P < 0.05).(5)Sishenwan high,medium,low dose and Dicetel group can downregulate Ghrelin,GHSR protein expression of gastric and duodenum(P < 0.05,P <0.01),and Sishenwan high dose group is particularly evident.Ghrelin and GHSR expression of braingut in Sishenwan medium dose group showed no significant difference with Dicetel group.Conclusion:(1)sishenwan can improve the diarrhea and gastrointestinal motility in Asdthenic Splenonephro-yang diarrhea model rats,and inhibit the intestine movement;(2)Sishen wan in the treatment of Asdthenic Splenonephro-yang diarrhea are dose dependent,Sishen wan high dose group has best effect;(3)the incidence of Asdthenic Splenonephro-yang diarrhea model rats may be associated with the up-regulation of Ghrelin,GHSR protein and gene overexpression.(4)The mechanism of Sishenwan on Asdthenic Splenonephro-yang diarrhea model rats may be associated with the down-regulation of Ghrelin,GHSR protein and gene overexpression.
