ObjectiveOn the basis of pre-study further explore the pathogenesis of early knee osteoarthritis(KOA),studies the mechanism of Guizhou Miao fumigation therapy treatment of early osteoarthritis in rabbits,to observe the effect of the expression of early knee osteoarthritis of rabbit cartilage tissue factor protein.MethodsThis experiment takes the New Zealand rabbit as the research object.The early KOA model was prepared by the method of injecting the rabbit knee joint into the cavity.We take Guizhou Miao fumigation prescription for fumigation treatment,and with clear water fumigation treatment as control,after 20 days of treatment,observe articular cartilage tissue pathological changes by gross observation and light microscope observation,and Mankin' s scores are observed;The protein expression of Bax,TNF alpha,Bcl-2 and IL-1 are detected by Western blot(blot Western).Results(1)The gross observation results can be seen in degenerative changes in the model group articular cartilage surface,Miao medicine fumigation group joint surface cartilage degenerative changes compared with the model group reduced.(2)Light microscope observation:Blank group:normal cartilage thickness,four layers structure clear,the tide line clear,complete structure;Cartilage surface smooth,level off;Pit distribution uniform normal cartilage cells,cartilage,cartilage matrix normal dyeing.Model group:thinning of the cartilage thickness,four layers structure level is disorder,the tide line unclear,incomplete structure;Cartilage surface erosion,fibrosis,radiation layer and deep cartilage matrix fiber loose,fat deposits;Surface and radiation layer of cartilage cells decreased significantly,the deeper the cluster cartilage cells can be increased,cell arranged disorder,normal cartilage pit disappear;Cartilage matrix dyeing severely reduced.Miao medicine fumigation groups:close to normal cartilage thickness,four layers structure layer is clear,the tide line is clear,but the structure is not complete;Cartilage surface rough;Surface of fibrosis,radiation layer and deep cartilage matrix fibres is normal;Cartilage cells with normal cartilage,but disordered arrangement of cartilage cells and deep cartilage cells of the cluster,normal cartilage pit part disappear;Cartilage matrix dyeing slightly reduced.Water control group:thinning of the cartilage thickness,four layers structure level is disorder,the tide line unclear,incomplete structure;Radiation layer and deep cartilage matrix fiber loose,a small amount of fat deposits;Surface layer of the decrease in the number of chondrocytes,and radiation deep visible cartilage cells of the cluster,disordered arrangement,normal cartilage pit disappear;Cartilage matrix dyeing severely reduced.(3)Mankin 's score results show:the model group increases significantly compared with the blank group score,with significant difference(P<0.01);Miao medicine fumigation group compares with model group scores declined,with significant difference(P<0.01);Water control group compares with model group score decreased,with statistical difference(P<0.05);Miao medicine fumigation group score decreases compared with clear water controls,with statistical difference(P<0.05).(4)western blot test results show:the model group compared with blank group comparison IL-1?TNF alpha?Bax protein expression levels increased significantly,the difference was statistically significant(P<0.05);Miao medicine fumigation group compared with model group,IL-1?TNF alpha?Bax protein expression level decreased,the difference was statistically significant(P<0.05);Miao medicine fumigation relatively clear water controls IL-1,TNF alpha?Bax expression decreased,the difference was statistically significant(P<0.05).Model group compared with blank group to compare the Bcl-2 protein expression level decreased obviously,the difference was statistically significant(P<0.05);Miao medicine fumigation group compared with model group the Bcl-2 protein expression increased obviously,the difference was statistically significant(P<0.05);Miao medicine fumigation relatively clear water controls the Bcl-2 protein expression levels drop,the difference was statistically significant(P<0.05).Conclusion(1)IL-1?TNF alpha?Bax?Bcl-2 factor is closely related to the pathological process of knee osteoarthritis.(2)The experimental model is successfully prepared through the general macroscopic observation,light microscope observation,Markin' s score and the correlative factors of protein expression.(3)This experiment confirmed that in the KOA articular cartilage of IL-1,TNF-alpha,Bax protein expression increased,Bcl-2 protein expression decreased,may be part of the pathogenesis of KOA.(4)By western blot test results obtained,Guizhou miao fumigation therapy can effectively control the IL-1,TNF alpha,Bax,Bcl-2 factor protein expression,showed the miao fumigation therapy to early prevention and treatment of KOA plays an important role.
