Objective:To observe theeffectofBDNFmRNA onthe expressionof miRNA-206-3p in C6 rat glioma cells affected by A?and to explore the anti-dementia effect of bushentiansui formula and its possible mechanism.Theoretical and experimental basis are provided to guide the clinical application and development of new drugs.Methods:1.Preparation of serum containing serum:Healthy SD rats,were randomly divided into control group and Bushentiansui formula group,animals in each group were given the corresponding drug or saline.The concentration of the rats in the low and high dose group was 1:6,after taking blood,centrifuge spare..2.C6 cell culture:C6 cells were cultured,the logarithmic growth phase cells were studied,treatment of Alzheimer's disease cell model with?-amyloid protein(A?25-35).Divided into control group,model group,low doze BSTSF group,high doze BSTSF group.3.The different expression of BDNF mRNA and miRNA-206-3p mRNA were observed by real-time fluorescence quantitative PCR.4.Western Blot was used to detect the expression of BDNF protein.Results:1.To comparison the real-time quantitative PCR reaction of BDNF mRNA,miRNA-206-3p mRNACompared with the control group,the levels of BDNF mRNA in the model group were significantly decreased?P<0.01?;Compared with the model group,the BDNF mRNA expression each of the Bushentiansui formula serum groups was significantly increased?P<0.05,P<0.01?.Compared with the control group,the levels of miRNA-206-3p mRNA in the model group were significantly increased?P<0.01?;Compared with the model group,the miRNA-206-3p mRNA expression each of the Bushentiansui formula serum groups was significantly decreased?P<0.05,P<0.01?.3.Western Blot Detection of BDNF Protein Results:Compared with the control group,the levels of BDNF protein in the model group were significantly decreased?P<0.01?;Compared with the model group,the BDNF protein expression each of the Bushentiansui formula serum groups was significantly increased?P<0.05,P<0.01?.Conclusions:1.The expression of BDNF protein and BDNF mRNA in AD cell model decreased and the expression level of miRNA-206-3p mRNA increased.2.BushentiansuiformulaintervenesafterADcellmodel,miRNA-206-3p mRNA expression levels decreased,BDNF protein?BDNF mRNA expression levels increased.Suggesting that Bushentiansui formula may be down by miRNA-206-3p mRNA,increased BDNF mRNA to increase BDNF protein expression.
