Effects Of Bushen Yizhi Formula Extracts On Learning And Memory Capacity And Synaptic ProteinsExpression In Alzheimer's Disease Mice

ObjectiveTo explore the effect of Bushen Yizhi Formula extracts(BSYZ)on learning-memory function and the expression of SYN,PSD95 and PSD93 in Alzheimer' s disease(AD)mice.Methods1.7 month male SAMP8 were devided into model group,donepezil positive group,and low-dose,middle-dose and high-dose BSYZ groups randomly.In addition,13 male SAMR1 with same age were used as negetive controls.2.The negetive group and model group were treated with distilled water,BSYZ groups treated with BSYZ by dose of 2.92 g/kg per day,5.84 g/kg per day,11.68 g/kg per day,and donepezil group treated with donepezil by dose of 3mg/kg per day.3.Groups of mice were observed by Morris water maze changes in its learning-memory function.The HE staining was used for the observation of the histopathological changes in the hippocampus,and the protein expression levels of SYN,PSD95 and PSD93 in the brain tissues were detected by Western blotting.ResultsEscape latency of mice in the model group was prolonged,neuronal damage in the hippocampus tissues was obvious,SYN,PSD95 and PSD93 protein expression in the brain tissue were decreased compared with the control group.Compared with the model group,the escape latency in middle-dose BSYZ group was significantly shortened.SYN,PSD95 and PSD93 protein expression in the brain tissue were increased,neuronal damage in the hippocampus tissues was significantly improved in all medication groups.ConclusionBSYZ can improve learning-memory function in AD mice probably by up-regulating the protein expression levels of SYN,PSD95 and PSD93 in the brain tissues.
