Alzheimer's disease(AD)is a neurodegenerative disease.Most AD risks remain largely unknown until now.MicroRNAs(miRNAs)are known as key post-transcriptional regulators of gene expression,and usually achieve function by its sequence specificity.More and more evidence has shown that microRNAs are involved in development of nervous disease and associated with AD risk.Here,we performed a systematic analysis to identify the potential miRNAs involved in AD risk.In stage 1,we performed a genome-wide mi RNA-based association study using the large-scale AD GWAS dataset,and successfully identified 6 significant miRNAs with P < 0.05.In stage 2,we conducted an AD case-control mi RNA expression analysis,and found only the miR-218-5p to show significant up-regulation in AD cases compared with normal controls.In stage 3,we predicted the target genes of miR-218-5p,and then performed an AD case-control gene expression analysis,and found the target genes of mi R-218-5p are significantly differently expressed in AD brain.In stage 4,we conducted a pathway analysis for miR-218-5p target genes,and identified that the target genes of miR-218-5p were significantly enriched in kinds of pathways related to nervous system.In summary,all these findings indicate that miR-218-5p is associated with AD risk,and miR-218-5p is a key regulator in nervous system.We believe that these findings provide important information about the molecular networks that miR-218-5p affects gene expression in AD risk.
