Thesium chinense, all grass sandalwood plants of Thesium chinensis, it has many another name just as incarnata, it distributed in southern and Northern china Widely. It has pungent smell, taste bitter slightly, it can clear heat and removing toxicity, and it can invigorating kidney and nourishing essence, can cure acute mastitis, pneumonia, lung abscess, tonsillitis, upper respiratory tract infection.Thesium chinense is perennial and parasitic herb, it has parasitic roots in root, it attached to other living plant roots and growth. Studies show that flavonoids are the main active substances of the Thesium chinense. Flavonoid is a kind of bioactive substances with a wide range of biological activities, such as anti-oxidation, anti-tumor, anti-virus, prevention and treatment of cardiovascular diseases and enhance human immunity and so on. The mainly flavonoid compounds in Thesium chinense are kaempferol glycosides, quercetin, isorhamnetin grain alcohol glycosides and so on, which kaempferol glycoside content is the highest, quercitrin, isorhamnetin glycosides content is less, so in this article, we mainly to research the process of extraction and purification of Thesium kaempferol glycosides.Kaempferin is a flavonoid glycosides compounds exist in Bairui grass, galangal, Astragalus complanatus, Ginkgo biloba and other traditional Chinese medicine commonly. It has medicinal activities of antioxidant, anti-inflammatory, anti-radiation and the prevention of cardiovascular disease and other pharmacological. The purification and separation of kaempferol glucoside component in kaempferin is few reports related to domestic. Therefore, the new technology of the separate and purificate kaempferin in Thesium chinense can make a a certain significance to the development utilization of Thesium chinense.1. In this paper, it establish a method for the content determination of flavonoid aglycones in Thesium chinense Turcz and its extracts. Due to there are many kinds of flavone glycosides in Thesium chinense, the component is complex, if determined all flavonol glycosides respectively, not only the workload is huge, and there is no significance for the practical. Consider the flavonol glycosides has a common aglycone of kaempferol, so we let the extraction of flavonol glycosides hydrolysis to kaempferol, to determination the content of flavonol glycosides in Thesium chinense Turcz. The grass extract solution is methanol?water ? hydrochloric acid(7:2:1), let it hydrolyze 3h under at 68?, and then detect the content of flavonoids in its hydrolysate. The chromatographic condition is: the flow phase was methanol?water? phosphoric acid(48:52:0.3), flow rate is 0.8 mL/min-1; wavelength is 254nm; column temperature is 45?. The results showed that the method was simple, rapid,accurate, high sensitivity and good reproducibility.2. By using different drying methods of dry in the shade, dried in the sun, and different drying temperature treat to fresh Thesium chinense, determination the content of kaempferol glycosides, shows the effect of different drying methods on the content of effective components, and study the influence of mildew to content of kaempferol glycoside. Using different drying methods treat to Thesium chinense Turcz., results show that, Two kinds of flavonol glycoside content change is kiln drying = dried in sun > dry in the shade > enzyme, which fixing in 105 ? and then dried in 80 ? and the dried in 80? is the highest, the drying time of dried in the shade is relatively long, so the content of flavonol glycoside is reduced, and the content of the sample through mildew and dried in 80 ? was the lowest. It can be seen that drying and drying in sun can be used to effectively maintain the content of two kinds of flavonoids in Thesium chinense Turcz.3. Through the selection of extraction parameters of solvent, temperature, time and material liquid ratio, determined the optimum extraction conditions were: the extraction temperature was 80 ?, the ratio of material to liquid was 1:20, the extraction time was 2.5h. The results show that the method is simple, rapid and effective.4. Select five macroporous resin of HPD-417, polyamide, DM130, AB-8 and F8 use to make experiments of extract static adsorption and desorption of Thesium chinense Turcz, determined using polyamide to purification of Thesium chinense Turcz extract liquid of flavonol glycosides. Through single factor screening test of the dynamic adsorption of polyamide purification conditions and then determined the polyamide optimum purification conditions of flavonol glycosides in Thesium chinense Turcz was: diameter ratio of is 1:10, elution is 60% ethanol, elution solvent volume is two times of column volume. After purification, the purity of kaempferol glycosides could reach 70%, the yield was 92%, the recovery rate was 99.8%.5. According to the pharmacological effects of kaempferol glycosides, this paper take the activity preliminary screening of kaempferol glycosides. The results showed that the Thesium kaempferol glycosides has hypoglycemic effect,but the effect of antiviral is not obvious.The reaserch show that the process of the extraction and purification used in this paper is stable and reliable, the method of determinate content is accurate and reliable, and the whole process system is simple and suitable for pilot scale production, it achieved the anticipated goal of this research.
