Objective To investigate the effect of transplantation of Spheroid Cultured Mesenchymal Stem Cells on the neurological function and expression of Nogo-A and NgR after cerebral ischemia-reperfusion injury in rats.To discuss the possible nerve protective mechanisms of stem cells in rats with Mesenchymal Stem Cells after ischemia. To provides a theoretical basis for the treatment of cerebral ischemic injury.Method The experimental animals were randomly divided into Sham group, Vehicle group, MSCs treated group. The focal cerebral ischemia and reperfusion model was induced by ligation with nylon monofilament in rats.Unplugging the fishing line for reperfusion in 2h after ischemia.MSCs were transplanted in MSCs group and mediums were given to the Vehicle group in 1d.On the1 st day,3rd day,7th day afer transplantation,using the method of RT-PCR to detect the expression of Nogo-A and NgR m RNA,the method of Western blot to detect the expression of Nogo-Aand NgR protein.Results1.MSCs labeled express in the MSCs group,not in the Vehicle group;2.Nissl's staining shows that there are more Nissl body in MSCs group than Vehicle group;3.Compared with the Vehicle group,neurological function were improved in MSCs group on the 7th day(P<0.05);4.Compared with the Vehicle group,the expression of m RNA of Nogo-A and NgR were down-regulated significantly on the 1st,3rd and 7th day(P<0.05);5.Compared with the Vehicle group,the expression of protein of Nogo-A and NgR were down-regulated significantly on the 1st,3rd and 7th day(P<0.05).Conclusion MSCs has neuroprotective effects and its mechanism may be associated with decresing brain tissue Nogo-A and NgR content.
