ObjectiveTo seek the effect of PTS on expression of BDNF and Nogo-A at different time after MCAO(middle cerebral artery occlusion)and the function of PTS on synapse regeneration in rats.MethodsCreate a MCAO model: male SD rat(w=300±20) were used in this study, improve Longa method cause middle cerebral artery occlusion. The neurological deficit grades of cerebral infarction were estimated by Longa method. After operation, rats were randomly divided to 3 groups: model(treat with N.S), TCM group(PTS) and Western medicine group(nimodipine), according to Longa and weight. N.S,PTS and nimodipine were intragastric administration once day and lasted 3d, 7d and 28d. Nogo-A,BDNF,TrkB,SYN and PSD-95 in cortex were observed, and the IL-6 expression were estimated by biochemical criterions. Before drawing the materials from rats, we have gathered tongue demonstration by figure camera.Results:1,The results of whole condition:After operation 3d, the Longa scores significant low before drug self-administration in PTS group and nimodipine group, the N. S group had no different in that. Weight: 3days after MCAO, all the treating groups and model groups weight have decreased. Tongue colour: After operation 3 days, all the Tongue colour of treating groups compared with normal group were bad. After operation 7d, the neurological function of both the treating groups and model group got close to the normal level. The weights of all the groups were increasing except for model group. The tongue colour of PTS group approach normal group, and outweigh model group. After operation 28d, Both the treating groups and model group have no neurologic impairment. The weight of all groups increased, and PTS group got close to nomal groups at the increasing extent. There were no statistic different between PTS group and normal group at the tongue colour2,Effect of PTS on expression of SYN and PSD-95 at different time after MCAO: Compared with the normal group, the expression of SYN was significant drop at the treating groups and model group after MCAO 3d and 7d. At 28d after MCAO, The expression of SYN got close to normal group at PTS and nimodipine group, and more than model group.3,Effect of PTS on expression of BDNF and TrkB at different time after MCAO:The cerebral infarction lead to the high expression of BDNF and TrkB, with time flying, the high expression gradually descend PTS group and nimodipine group were higher than model group at the 3 different time point.4,Effect of PTS on expression of Nogo-A at different time after MCAO:The experimental result show that the expression of Nogo-A of model group significantly increased at ealeier, at 28d, it is descend. PTS group and nimodipine group were lower than model group at the 3 different time point.5,Effect of PTS on IL-6 of serum at different time after MCAO:The level of IL-6 sharply raise, Following the course of disease, the level of serum IL-6 was descend. PTS group and nimodipine group were lower than model group at the 3 different time point.Conclusion:1. PTS can improve the whole condition and colour of tongue of MCAO rat, and reduce the neurologic deficiency.2. PTS can enhance the synaptic remodeling after MCAO.3. PTS can be up-regulate the expression of BDNF and TrkB after MCAO.4. PTS can be down-regulate the expression of Nogo-A after MCAO.5. PTS can make the level of serum IL-6 lower after MCAO6. PTS can increase the expression of BDNF and TrkB, decrease the expression of Nogo-A, promote the expression of SYN and PSD-95 in rat brain after middle cerebral artery occlusion, which may be the one of remodeling mechanisms of PTS.
