Objective: To compare the expression level of ?7n Ach R,TNF-? and HMGB1 in a guinea pig model of acute cholecystitis by electroacupuncture at Yanglingquan, Shangjuxu, Xiajuxu, Zusanli points, and to explore whether Yanglingquan has relative effective specificity when treating its corresponding viscera disease.Methods: seventy-two(50% male and 50% female) SD guinea pigs were divided randomly into 6 groups : Blank group, Model group, Yanglingquan group, Shangjuxu group, Xiajuxu group, Zusanli group. The AC guinea pig model was made by injection of Escherichia coli into the gallbladder. Then the six treatment groups respectively received electroacupuncture with its corresponding points for 5 consecutive days. After treatment, the gallbladder pathological tissue was observed by HE staining, the level of ?7n Ach R in gallbladder tissue was detected by Western blot, the TNF-? expression in gallbladder tissue was observed by immunohistochemistry, and HMGB1 level in serum was checked by ELISA.Results: 1 Observation of gallbladder morphology under light microscope(10X40 times), The results showed that the blank group: Monolayer columnar epithelium cells were arranged in neat rows in mucosal epithelial tissue. The inherent film is thin, containing part of the small blood vessels and lymphatic vessels. There was no dilatation and congestion in the lamina propria and tunica submucosa. The grassroots arranged irregularly, and there was no inflammation reaction in the integrated plasma membrane. Model group: There was a large number of mucosal epithelial necrosis, hyperemia and edema, and there was a large amount of inflammatory cell infiltrating into the lamina propria. Yanglingquan group: The mucosal epithelial remained largely intact. The hyperemia and edema existed in the loose lamina propria, with infiltration of a small amount of lymphocytes and neutrophil. There was a little congestion in muscular layer and serous layer. Other points group: There was a small defect in mucosal epithelium. There existed obvious vascular dilatation and congestion in the inherent layer and submucosa and muscular layer with neutrophil infiltration.2. The expression of gallbladder tissue ?7nAchR among each groups: Compared with normal group, its ?7n Ach R level were obviously decreased(P<0.01) in other groups. When compared with model group, the level of ?7n Ach R in four point groups were signifcantly increased after acupuncture treatment(P<0.01). And a further comparison of the four point groups, the level of ?n Ach R in other groups was decreased compared with Yanglingquan group(P<0.01 or P<0.05).3. The expression of serum HMGB1 among each groups: Compared with normal group, the serum level of HMGB1 were obviously increased in other groups(P<0.01). When compared with model group, the level of HMGB1 in four point groups were signifcantly decreased after acupuncture treatment(P<0.01). And a further comparison of the four point groups, the level of HMGB1 in other 3 groups was increased compared with Yanglingquan group(P<0.01).4. The expression of TNF-? in gallbladder tissue: Compared with normal group, its TNF-? level were obviously decreased in other groups(P<0.01). When compared with model group, the level of TNF-? in four point groups were signifcantly increased after acupuncture treatment(P<0.01). And a further comparison of the four point groups, the level of TNF-? in other groups was increased compared with Yanglingquan group(P<0.01 or P<0.05).Conclusions: 1. EA can control the inflammation reaction in AC by regulating the expression of TNF-?, ?7n Ach R and HMGB1, which can reduce the inflammatory reaction and protect gallbladder tissue from the inflammation injury by suppressing the inflammatory factors.2. The treatment effect of Yanglingquan points were better than the others points, which partly proved that Yanglingquan has relative specificity when treating its corresponding viscera disease, namely, AC...
