Objective:To compara the expression level of IL-1?,NF-?B,HMGB1 in acute cholecystitis model guinea pig by electroacupuncture at Yanglingquan,Shangjuxu, Xiajuxu,Zusanli points,and to explore whether Yanglingquan has relative specificity when treating its corresponding viscera disease.Methods:72 healthy guinea pigs(50%male and 50%female)were randomly divided into 6 groups:a blank group,a model group,Yanglingquan group,Zusanli group,Shang juxu group and Xiajuxu group,12 guinea pigs in a group.Set up the acute cholecystitis guinea pigs models through injection of Escherichia coli at the right buttock.Then the six treatment groups respectively recived electroacupuncture with its corresponding points for 5 consecutive days.After treatment,abdominal aorta blood sampling in guinea pigs and gallbladder tissue was stained with HE.Changes of gallbladder morphology in guinea pigs were observed,ELISA is used to check models'serum IL-1? level and the expression of NF-kB and HMGB1 in the gallbladder were detected by immunohistochemistry.Results:1 Observation of gallbladder morphology under light microscope. The results showed that there was no pathological change in the gallbladder in the blank group;in model group, a large number of inflammatory cells were found and ulcer formation;Yanglingquan, Zusanli, Shangjuxu, xiajuxu group of guinea pig gallbladder mucosa pathological injury was alleviated and Yanglingquan group is more obvious.2. The expression of gallbladder tissue NF-?B among each groups:compared with blank group, its NF-?B level were obviously decreased(P<0.01);after acupuncture treatment, when compared with model group, the level of NF-?B inYanglingquan groups were signifcantly increased(P<0.01), While the rest with point group only had an decreased trend but no statistically significant(P>0.05).When compared with Yanglingquan group.the tissue NF-kB level of Zusanli group,Shangjuxu group and Xiajuxu group were all increased with statistically significant difference(P<0.01 or P<0.05).3. The expression of gallbladder tissue HMGB1 among each groups:compared with the blank group, the expression level of HMGB1 was significantly higher in the gallbladder of guinea pigs.After acupuncture treatment,when compared with model group, HMGB1 of Yanglingquan group was signifcantly decreased(P<0.01). While the rest with point group only had an decreased trend but no statistically significant (P>0.05).When compared with Yanglingquan group, the tissue HMGB1 of other three acupoint groups were all improved increased.(P<0.01 or P<0.05).4.The expression of gallbladder serum IL-1? among each groups:compared with the blank group.its IL-1?level were obviously increased(P<0.01).compared with the model group,the serum IL-1?were all decreased with statistically significant difference(P<0.01 or P<0.05);compared with Yanglingquan group,the serumIL-1? level of Zusanli group, Shangjuxu group and Xiajuxu group were all increased with statistically significant difference(P<0.01 or P<0.05).Conclusions:lElectroacupuncture at Yanglingquan,Zusanli, Shangjuxu and Xiajuxu all could significantly decrease the inflammatory reaction in guinea pigs.It may be achieved by adjusting the factors related to IL-1??NF-kB and HMGB1 signaling pathway,regulating inflammatory response and improving pathological changes of gallbladder tissue.2.Comparison shows that the intervention effect of Yanglingquan points were better than others points in guinea pigs acute cholecystitis models,which partly proved that Yanglingquan point treatment of acute cholecystitishas relative specificity.3.This experiment's results partly confirmed that "curing viscera diseases by He points",refers to that the clinical significance of lower He-sea points lies mainly intreating its corresponding viscera's disease,its specific mechanism still need further study.
