Comparative Study Of Electroacupuncture On Shangjuxu And Quchi For Treating Ulcerative Colitis In Rats--Tentative Exploration On "Curing Viscera Diseases By He Points"

Objectives: To compare the acupuncture effect of Shangjuxu point(lower He point of large intestine Fu) and Quchi point(He point of large intestine meridian) on treating ulcerative colitis(UC) on rat. To get the experimental evidence of the theory of "curing viscera diseases by He points" and academic guidance on clinical practice.Methods: Forty male Wistar rats were randomly assigned into four groups: Normal group(A), Model group(B), Shangjuxu point group(C), Quchi point group (D), each group has ten rats. Each colon tissue of these rats was investigated on view of naked eyes or adding a magnifying mirror and microscope. The active value of MPO, the content of IL-lβ VIP and expression of IL-lβmRNA of colon mucosa in every group were also examined.Results: 1. Morphological change of UC rats in every group: 0 Observation by unaided eye and evaluation: it was smooth light, clean inA's colon tissue; there existed abroad erosion, and some UC foci in B; the main problem in colon tissue of C is hyperemia or edema, while erosion, superficial focus could also seen in few rats; there existed obvious erosion multi-ulcer-focus in D. On aspect of ulcer index(UI) there was an obvious difference between B and C (P<0.01) or between C and D (P<0.01) , no distinct difference between C and B(P>0.05). (D Obsevation by microscope and classification: normal colon tissues in group A; necrotic and fibrosis foci of mucous layer and tela submucosa in group B; main manifestation in treatment group: inflammatory infiltration in tela submucosa, some superficial erosion and punctate ulcer in group C while necrotic mucous layer and inflammatory infiltration in group D. There were significant differences between C and B(P<0.01) or between C and D(P<0.01), but not between group C and B (P>0.05). 2. Comparison of MPO activity in tissue of colon mucosa of rats of UC: compared with B, MPO activity in C decrease obviously (PO.05); there was a closely comparation between D and B(P>0.05); there was an obvious difference between D and C(P<0.05). 3. Content of IL-1 of colon mucosa: compared with B, content of IL-1 of colon mucosa in C decrease obviously (PO.05); there was a closely comparation between D and D(P>0.05), and that of D was higher distictly than that of C, but it has BO statistics significance(P>0.05). 4. Expression of IL-l mRNA in the colon mucosa: compared with B, expression of IL-lmRNA in C decrease obviously (PO.05); there was also a clear comparation between D and B(P<0.05) and between D and C(P<0.05). 5.Content of VIP in the colon mucosa: compared with B, expression of IL-lpmRNA in C decrease obviously (.P<0.05); there was also a clear comparation between D and B(P<0.05) but a closely comparation between D and C (P>0.05).Conclusions: 1. Comparing with the morphological change of UC rats in every group and the value of MPO, electroacupuncture on Shangjuxu point have a significant effect on UC, it indicates its relative correlation with large intesine fu. 2. Electroacupuncture on Shangjuxu point could inhibit expression of IL-l[3mRNA, decrease promotion of inflammation of IL-lf3. It probably is one of the important mechanisms of electroacupuncture on Shangjuxu point to treat UC. 3. Electroacupuncture on Shangjuxu point could decrease the content of tissue VIP , it indicates that influencing the content of tissue VIP is also probably an important adjusting mechanism. 4. Comparing the effect and the mechanism of electroacupuncture on Shangjuxu and Quchi for treating ulcerative colitis in rats , electroacupuncture on Shangjuxu point had more significally difference of treating UC than Quchi point, it indicates that "curing viscera diseases by He points" theory of treating viscera diseases by lower He point has its scientific content. It needs study further to reveal the whole content of "curing viscera diseases by He points" theory.
