Purpose:Rhizoma Drynariae with kidney strong bone,continued to hurt the analgesic effect,is one of the traditional Chinese medicine Chinese medicine orthopedics and traumatology,the most commonly used early in the Song Dynasty,Rhizoma Drynariae was used for the treatment of“tendon rheumatism”,namely the modern medicineosteoarthritis. Osteoarthritis is characterized by the degeneration of articular cartilage of chronic diseases. To explore the clinical experience of Rhizoma Drynariae,based on prior treatment of the disease related to the medical fieldexperiment,this study by TUNEL(terminal deoxynucleotidyl transferase)and its effect on the apoptosis of synovial cells of rabbit osteoarthritis of knee joint detection method,Western-blot(Western blot)method was used to detect the different expression of Fas?Fasl protein in synovial cell apoptosis factor,preliminary study the mechanism of action of Rhizoma Drynariae on apoptosis of synovial cells of rabbit knee osteoarthritis,for further clinical application and promotion of Rhizoma Drynariae,and to provide basis for the research and development of new drugs. Material and method:In 24 New Zealand white rabbits were randomly divided into normal group,model group,treatment group and dimensional force of Rhizoma Drynariae group,each group 6 only. The left hind limb extension using animal model method of tube type gypsum fixedcause of osteoarthritis of the knee,fixed for 6 weeks.After the success of modeling,the treatment group according to 1.5g/Kg/d dosage by intragastric administration of Rhizoma Drynariae,control group according to 7Omg/Kg/d dosage by intragastric administration dimension force,model group and normal group were given normal saline by gastric feeding the same dose. After 4 weeks of treatment, all animal use ear marginal venous air embolism method executed,remove the left hind limb of each rabbit articular cartilage of the knee joint synovium,isolated. Part of a fixed in formalin solution,used to observe the morphological changes of synovial cell in HE staining under light microscope,detection of synovial cell apoptosis by TUNEL;the other part of detection of Fas?Fasl protein expression level of Western-Blot method. The implementation of statistical analysis of experimental data using SPSS15.0 statistical software,with P<0.05 indicates statistical significance. Results: 1.Rabbit knee OA model 4 weeks of synovial cell apoptosisphenomenon,apoptosis of cell TUNEL in synovial tissue staining was positive in the normal articular,only weak positive expression,model group of TUNEL positive synovial cells quantity,significantly higher than that in the normal group(P<O.01).Rhizoma Drynariae treatment group,control groupthe number of dimension force of synovial cells in TUNEL positive markers are more,higher than the normal group(P<O.01),significantly lower than that in the model group,the differences were statistically significant(P<O.01),illustrated the application of Rhizoma Drynariae,dimensional force can reduce the apoptosis of rabbit knee osteoarthritis synovial cells. Rhizoma Drynariae treatment group,dimensional force controlgroups,there is significant difference(P<O.01),description of Rhizoma Drynariae reduce apoptosis effect is superior to that of dimensional force of rabbit knee osteoarthritis model synovial cells. 2.Rabbit knee OA model 4 weeks of synovial joints of Fas?Fasl protein expression level is not the same. The normal group Fas,model group showed weak expression of FasL,Fas?FasL showed strong expression,the difference between the two groups were statistically significant(P<O.01).Treatment group,Rhizoma Drynariae dimension force can be reduced Fas,the expression of FasL,its protein expression level is higher than the normal group(P<O.01),lower than that of model group(P<O.01). Comparison between the two groups,there was a significant difference between the expression of Rhizoma Drynariae,explains Fas?FasL in cut protein levels and external force strong. 3.After 4 weeks,synovial pathological changes of osteoarthritis,Rhizoma Drynariae,dimensional force after treatment of synovial ultrastructure had a bit of improvement,can play a protectiverole. Conclusion: 1.Excessive apoptosis Drynariae can inhibit osteoarthritis synovial cells. 2.Fas?FasL,enhanced expression of protein in synovial cells in osteoarthritis,its pathogenesis is closely related to.The expression level of Rhizoma Drynariae through down-regulation ofosteoarthritis of knee Fas?FasL Protein,relieve osteoarthritis synovial cell apoptosis. 3.Rhizoma Drynariae on apoptosis of synoviocytes in osteoarthritis of the knee joint is better than the dimensional force effect.
