Objective: To establish a model of osteoarthritis of knee in New Zealand white rabbits and to observe the effect of total flavonoids from rhizoma drynariae on IL-1?,MMP-3 and MMP-13 in knee osteoarthritis of rabbits at different pathological stages.To explore the mechanism of total flavonoids from rhizoma drynariae in the treatment of osteoarthritis,and to provide theoretical basis for clinical treatment of osteoarthritis.Methods: A total of 56 male New Zealand white rabbits weighing about 2.5 kg were randomly divided into 7 groups.7 groups were normal group,early medication group,early model group,medium term medication group,medium term model group,late medication group,late model group,8 rabbits in each group.In the model group and the medication group,the knee osteoarthritis model was established according to the modified hulth method and the normal group was not treated.In the medication group,the total flavonoids from rhizoma drynariae were given 8 weeks,and the rabbits in the model group were given normal saline for 8 weeks.The early,middle and late stages were given at 4,8 and 12 weeks after modeling.Each group was sacrificed after 8 weeks of intragastric administration.Articular cartilage was used for HE staining.The expression of IL-1? and MMP-3 in serum,synovial tissue and synovial fluid were detected by ELISA.The expression of MMP-13 in synovial tissue was detected by immunohistochemical staining.The data were analyzed by SPSS 17.0software.Results: Mankin's score: early,middle and late medication group was lower than theearly,middle and late model group(P <0.05).The concentration of IL-1? in serum,synovial tissue and synovial fluid : the early,middle and late medication groups were lower than those of the early,middle and late model groups(P <0.05).The concentration of MMP-3 in serum,synovial tissue and synovial fluid: the early,middle and late medication groups were lower than those of the early,middle and late model groups(P <0.05).The positive rate of MMP-13 in synovial tissue: the early,middle and late medication groups were smaller than those in the early,middle and late model groups(P <0.05).Conclusion: The total flavonoids from rhizoma drynariae had a significant effect on cartilage degeneration in early,middle and late stage of rabbit knee osteoarthritis.It may reduce the destruction of chondrocytes and depletion of extracellular matrix in knee osteoarthritis by reducing the expression of IL-1?,MMP-3,MMP-13.
