Objective:To explore the beta defense-2 (rBD2) mRNA expression in the peripheral blood and lung tissue of rats after tracheotomy, IL-2 content in peripheral blood,T cell subsets with lung tissue, sIgA content change rule in inalveolar lavage and rhizoma drynariae intervention effect on it.Methods:All of 72 SPF health male Sprague wrote (SD)-Dawley rats were divided into three groups:normal control group(A), Tracheotomy cannula lien model with Tracheotomy tube(B)and the model drug group(C). To prepare model with tracheotomy.Collecting specimen in 24h,72h,168h three times after the success of the model. Collection of peripheral blood, lung tissue and alveolar lavage fluid, Detection of beta defense-2 (rBD2) mRNA expression in peripheral blood and lung tissue, content of IL-2 in Peripheral blood,Lung tissue T cell subsets (CD3, CD4, CD8), and the sIgA level in Alveolar lavages..Results (1)The rBD2 mRNA expression quantity decreased obviously in Peripheral blood of rats after tracheotomy, especially at 24h,the degree of decline slightly improved in the 72h and 168h,with the intervention of Rhizoma drynariae,the expression of rBD2 can be improved significantly after tracheotomy in peripheral blood of rat,the improvement effect is enhanced with the increase of the time,but did not return to normal levels. (2)The IL-2 content did not change in peripheral blood of rats after tracheotomy,but with the intervention of Rhizoma drynariae, the content of IL-2 in peripheral blood of rats increased significantly after tracheotomy,the degree of improvement was proportional to the time with medication.(3)The rBD2 mRNA expression in the lung tissue was significantly increased at the 24h after tracheotomy, it starts to fall in 72h and decline to the lowest value inl68h;With Chinese medicine Rhizoma drynariae intervention, The rBD2 mRNA expression was obviously increased and up to the highest value at 72h, but the expression was slightly decreased in 168h. (4)The slgA content of rat in alveolar lavage fluid was increased in 24h and 72h two hours after tracheotomy,but it decreased at 168h. With Chinese medicine Rhizoma drynariae intervention,the sIgA content was increased in 24h and did not fall at 168h.(5)The content of CD3 in lung tissue was significantly decreased after tracheotomy,it reached the lowest value in 168h;with taking Rhizoma Drynariae,the content of CD3 appears to rise, reaching the maximum value in 72h, and it is slightly down at 168h. (6) The content of CD4 in lung tissue was transient increased and then decreased significantly after tracheotomy, it declined the lowest value in the 72h and slightly increased in 168h;with taking Rhizoma Drynariae,it starts to increase at 72h and reached the maximum value at 168h. (7)The CD8 levels in lung tissue increased significantly after tracheotomy, it reached the maximum at the 72h; with taking Rhizoma Drynariae, it begin to decline at the 72h and its reached the lowest value on 168h.Conclusion:Rhizoma drynariae can enhance or protect the immune function of rats after Tracheotomy, especially pulmonary local immunity,This immune enhancement effect of Rhizoma Drynariae, which it may be associated with the increase β- defensin -2 mRNA expression in peripheral blood and lung tissues,improve the IL-2 content in peripheral blood,increase sIgA levels in alveolar lavage and improve T cell subsets imbalance effect for rats after tracheotomy...
