With the development of social scientific technology and the needs for exploration of the microscopic world,the digital microscope arises at the historic moment.Microscope image processing technology has been widely applied to the aspect of microprocess,such as industrial microprocess,biomedicine and historical heritage.Most of the methods used to measure objects are contact measurement,which can easily damage the surface of objects.Howere,it's not necessary to contact with the object by using microscopic visual measurement methods.It only needs to process the object images to complete non-contact measurement.Observing stereoscopic image of the object by microscope,the microprocess and measurement are realized by using the high-resolution stereoscopic image.Before the micro-precision processing is carried out,obtaining accurate focus binocular microscope image is guaranteed by the subsequent image processing.At present,most of the micro-measurement technology is from a slightly different angle of two fixed observation object.Then,it can obtain the depth of the object information,which is taken by the double channels on the same scene.While,the principle of imaging system is complex,which can not be freely set up the angle and is not easy to obtain satisfactory viewing effect.In this paper,the image processing technology of digital microscope were mainly researched.Basde on the focusing problem of binocular images,one judging method is proposed to determine the focusing consistency of digital stereo microscope.Then,based on the complicated structure of micro-measurement system,single objective stereo microscopic measurement method is proposed,which is based on LED illumination.For microscope stereo image clarity may be different at the same location,a method for determined the focusing consistency of stereo optical microscope is proposed.First,dot centers of different depth calibration board are extracted with region grown segmentation algorithm and regions to be evaluated are selected.Second,focus measure function is used to evaluate the selected image regions and definition-depth curve is obtained.Then the curve peak is got by curve fitting technique,the depth is draw and 3D depth measurement is completed.Finally,using the left and right eye dots in the centers of three-dimensional coordinates are fitted around the focal plane.Focus plane angle measured and distance of dot center focusing are used to analysis the focusability.Experimental results show that the proposed method can effectively determine the focusing consistency of stereo optical microscope.Basde on the complicated structure of micro-measurement system,single objective stereo microscopic measurement method based on LED illumination is proposed.The proposed method uses the principle of microscopic imaging to construct a measurement model which is easy to work,according to calibrating relationship between image offset and height of motorized translation stage,from which the height of object can be obtained.Experimental results demonstrate that the proposed method's measurement accuracy is 98.14~99.75%,which indicates that the proposed method can be used to measure height of micro object within tolerance error.
