Currently, high-tech research microscopes (such as the transmission electron microscope, scanning electron microscope) as optical instruments mostly have used auto-focusing system, and some microscopes are equipped with digital photography system. However, these equipments are expensive. This project aims to improve the ordinary microscopes and develop the digital acquisition system of the microscopic image which can accurately acquire the image information. It also can be wildly used in areas such as biomedicine, materials science, research experiments and so on.In this paper, we do some research about the image acquisition system of the digital microscope, and describe its design scheme. There are also descriptions about the detailed design of every part. The article has been discussed that it adopts step-by-step electric motor to come to auto-control through reforming the carrier and the objective lens, the design of the image acquisition device, from view systematic optical quality and demands to start off discussing to select and use CCD, adjustment mechanism of the Imaging lens is designed by adding the step-by-step electric motor pretending to realize high focusing accuracy and brings forward innovation design about the auto alignment device in micro field of view.The auto-focusing technology of acquiring microscopic image is studied, and an auto-focus algorithm is proposed to realize the microscopic image auto-focusing. The algorithm, which combines gray scale difference method and high-accuracy Laplacian function method, is able to properly select the focusing criterion function and step length according to the gradients of the focusing evaluation functions in each period of the focusing process. It utilizes a variable step-length method to realize the transition from fast rough adjustment to accurate one, which achieved satisfying results in practical uses. Image mosaic is an important research field of computer vision, image processing and computer graphic all the time. The method of image mosaic is generated while the critical technology such as image pre-processing, image registration and image composition is discussed according to the process of image mosaic. The method can significantly reduce the time of matching and solve the problem of joint.The digital acquisition system of microscopic image based on the exploitation of ordinary microscope has been successfully applied to the interactive teaching of remote medical treatment and the experiment of observing creatures by microscope with satisfying results.
