Effect Of Exercise Combined With Resveratrol On Vaspin MRNA,Protein Expression And Plasma Concentration In Aged Obese Rats

ObjectiveEffect of Vaspin mRNA and protein expression in visceral adipose tissue and plasma Vaspin concentration of aged obese rats were discussed through the intervention of different intensity exercise combined with resveratrol in 8 weeks.MethodsSelecting 6 healthy aging rats as the blank control group,selecting 36 aged obese rats were randomly divided to 6 groups,6 rats per group,e.g.the control obese group,resveratrol obese group,low intensity exercise obese group,and low intensity exercise combined with resveratrol obese group,and moderate intensity exercise obese group,and moderate intensity exercise combined with resveratrol obese group.The speed of the low intensity exercise was 12m/min,and the speed of the medium intensity exercise was 15m/min,15min/time,4 groups,rest between groups 5min,1h/d.Using 52.5mg/(kg·d)of resveratrol to fill the stomach,5times/week,8weeks of continuous intervention.After 8 weeks,Vaspin mRNA expression in visceral adipose tissue was measured by the qRT-PCR,Vaspin protein expression in visceral adipose tissue was measured by Western blot,plasma Vaspin and insulin concentrations were measured by ELISA,and blood glucose was measured by automatic biochemical analyzer,and insulin sensitivity was calculated.Results1.Effect of exercise combined with resveratrol on Vaspin mRNA expression in visceral adipose tissueVaspin mRNA expression in visceral adipose tissue of aged obese rats was significantly increased(P<0.01).Simply exercise,simply supplemented resveratrol and exercise combined with resveratrol could decrease its expression(P<0.01),it had less affected by exercise intensity.2.Effect of exercise combined with resveratrol on Vaspin protein expression in visceral adipose tissueVaspin protein expression in visceral adipose tissue of aged obese rats was significantly increased,its protein expression was decreased by low intensity exercise but it increased on moderate intensity exercise.The expression of Vaspin protein was reduced by simply supplemented resveratrol.Vaspin protein expression showed that it was increased by low intensity exercise combined with resveratrol but it was decreased by moderate intensity exercise combined with resveratrol.3.Effect of exercise combined with resveratrol on plasma Vaspin concentration, insulin sensitivity and blood glucose.Plasma Vaspin concentration and blood glucose were significantly increased in the aged obese rats(P<0.01),and insulin sensitivity was decreased(P<0.05).Blood glucose was decreased and insulin sensitivity was increased by exercise(P<0.05,P<0.01).Plasma Vaspin concentration had little effect by exercise(P>0.05).Blood glucose was decreased and insulin sensitivity was increased by exercise combined with resveratrol(P<0.05,P<0.01).Plasma Vaspin concentration had little effect by exercise combined with resveratrol(P>0.05).But the effect of exercise intensity wasn't different significantly.Conclusion(1)Vaspin mRNA expression in visceral adipose tissue of aged obese rats was significantly increased.Vaspin mRNA expression was decreased by both exercise and exercise combined with resveratrol.(2)Vaspin protein expression in visceral adipose tissue of aged obese rats was significantly increased,and the effect of exercise combined with resveratrol on Vaspin protein expression in adipose tissue of obese rats had no obvious regularity.(3)Plasma Vaspin concentration was increased in the aged obese rats,it was decreased by simply supplemented resveratrol.It had less effect by both exercise and exercise combined with resveratrol.(4)Insulin sensitivity was decreased and blood glucose was increased in aged obese rats.Insulin sensitivity was increased and blood glucose was decreased by both exercise and exercise combined with resveratrol,but the effect of exercise intensity wasn't different significantly.
