Objective: A SD rat obesity model was established.Eight weeks after successful modeling,aerobic exercise and vitamin D intervention were performed to detect nuclear factor-?B(NF-?B),monocyte chemoattractant protein-1(MCP-1),and tumor in adipose tissue.To explore the role of aerobic exercise and vitamin D in adipose tissue of adipose tissue in obese rats,and to explore the mechanism of inflammatory factors such as TNF-? and IL-6.Methods: Seventy SD rats were randomly divided into two groups: 10 in the normal diet group(C)and 60 in the high-fat diet group(HD).The group C was fed normal feed throughout,and the HD group was used to establish the obesity model and feed the high-fat diet.After the model was successfully established,they were randomly divided into 12 obese(O),12 obese+exercise(OE),13 obese + vitamin D(OV)and 13 obese + vitamin D + exercise groups(OVE).The aerobic exercise program is: treadmill exercise,speed 15 m/min(equivalent to 50% VO2 max load strength),each exercise 50 min,6d/w.Vitamin D gavage regimen: gavage dose of 5 ml/kg·BW,fasting before gavage,gavage once a day.All groups are free to drink water.At the end of the 8-week intervention,blood was taken from the heart to test the vitamin D content;epididymis and perirenal adipose tissue were isolated and morphologically studied by paraffin section HE staining.Adipose Tissue Inflammation Interleukin-6(IL-6),Monocyte Chemoattractant Protein-1(MCP-1),Nuclear factor-?B(NF-?B),Tumors were measured by double-antibody step-wise sandwich enzyme-linked immunosorbent assay(ELISA)The study of the molecular mechanism of TNF-? concentration.Results: 1.There was no significant difference in body weight between the groups before the experiment(P>0.05).After the model was established,the HD group was significantly higher than the C group(P<0.05);the Lee's index and the HD group were significantly higher than the C group.Increased sex(P<0.05).There was no significant difference in body weight and Lee's index between groups O,OE,OV and OVE before intervention(P>0.05).After intervention,compared with group C,body fat of rats in groups O and OV The body fat ratio was higher in the OE group and the OVE group than in the C group.The liposome ratio had no significant difference(P>0.05).Compared with the O group,the OE group and OVE There was a significant difference in the lipid body ratio of the rats(P<0.05).2.HE staining of adipose tissue revealed: Adipose tissue cells in group C were normal and there was no inflammatory cell infiltration.Compared with group C,adipose tissue cells in the O group,OV group,OE group and OVE group had different degrees of enlargement,there were different degrees of inflammatory cell infiltration phenomenon,and there was no obvious boundary between cells.Compared with group O,the size of adipose tissue in OV group was basically the same,and the infiltration of inflammatory cells was partially reduced;the adipose tissue cells in OE group were smaller,the infiltration of inflammatory cells was partially reduced,and the boundary between cells was clearer;OVE Adipose tissue cells in the section were near normal,with essentially no inflammatory cell infiltration and clear boundaries between cells.3.The content of IL-6,MCP-1,NF-?B and TNF-? in adipose tissue was significantly higher in group O than in group C((P<0.05);after 8 weeks of aerobic exercise and vitamin D intervention,OVE group The levels of IL-6,MCP-1,NF-?B and TNF-? in adipose tissue of rats were significantly lower than those in group O(P<0.01).The contents of IL-6 in adipose tissue of rats in OE group and OV group were significantly increased.Compared with group O,the levels of MCP-1,NF-?B and TNF-? in adipose tissue were significantly lower than those in group O(P<0.05);The contents of MCP-1,NF-?B and TNF-? in the tissue were lower than those in the O group,and there was no statistical significance(P>0.05).4.The content of vitamin D in HD group was significantly lower than that in group C(P<0.05);after 8 weeks of aerobic exercise and vitamin D intervention,the OVE group was increased compared with group O,with highly significant difference(P<0.01).The OV group was significantly higher than the O group(P<0.05).The OE group was lower than the O group,but there was no significant difference(P>0.05).Conclusion: 1.High-fat diets can increase body weight and lee's index in rats,activate inflammatory factors,and induce inflammatory reactions.2.Aerobic exercise and vitamin D combined intervention can activate anti-inflammatory factors,inhibit pro-inflammatory factors,reduce adipose tissue infiltration,inflammatory response;aerobic exercise intervention or vitamin D intervention has a certain degree of adipose tissue inflammation in obese rats The inhibitory effect of a combination of aerobic exercise and vitamin D on the inhibition of adipose tissue in obese rats is more pronounced.3.Simple obesity can lead to lower serum vitamin D levels,vitamin D supplementation can increase the serum vitamin D content,aerobic exercise and vitamin D combined intervention can effectively improve the utilization of vitamin D.
