Study On The Effect Of Ginseng Pectin WGPA On Improving The Exercise-induced Fatigue In Mice By Promoting Gluconeogenesis

With the development of modern competitive sports and national fitness,more attention has been paid to the recovery of exercise-induced fatigue.Ginseng pectin has a variety of biological activities,especially in anti-fatigue effect,but mechanism is unclear.WGPA was administered immediately after the mice were exhausted to investigate and analyze the possible mechanism of the effect of WGPA on recovery.In this study,a mice wheel fatigue apparatus was used,and C57BL/6J mice were subjected to exhaustive exercise.After exhaustive exercise,mice in group WGPA were fed with 200mg/kg of ginseng pectin WGPA immediately,the volume was 0.3mL,Control group was given the same volume of saline.After intragastric administration of fasting rest 3h,the serum,liver tissue and quadriceps were collected.The indexes included hepatic glycogen,muscle glycogen,blood glucose,blood lactic acid,lactate dehydrogenase in serum and liver,MDA,BUN,creatine kinase,the rate limiting enzyme activity and mRNA transcription level of hepatic gluconeogenesis were detected.The result showed that ginseng pectin WGPA can significantly reduced lactic acid,MDA and blood urea nitrogen levels in the serum compared with Control group(P<0.05),indicated that WGPA can promote the recovery of fatigue.Further study showed that WGPA can significantly increase the blood glucose(P<0.05),and promote the synthesis of hepatic glycogen(P<0.001),but had no effect on muscle glycogen(P>0.05).These suggested that the effect of WGPA may be related to gluconeogenesis.Gluconeogenic enzyme activity assay results showed that WGPA significantly up-regulated the activity of phosphoenolpyruvate carboxykinase(P<0.05)and glucose-6-phosphatase(P<0.01),while there was no effect on the activity of pyruvate carboxylase,fructose-1,6-bisphosphatase and lactate dehydrogenase in liver(P>0.05),indicating that WGPA promotes gluconeogenesis by improve the phosphoenolpyruvate carboxykinase and glucose-6-phosphatase activity.Further determination of their mRNA transcripts showed that WGPA significantly increased the mRNA level of glucose-6-phosphatase(P<0.01),but had no effect on the mRNA level of phosphoenolpyruvate carboxylase(P>0.05).In summary,ginseng pectin WGPA can promote gluconeogenesis to improve exercise-induced fatigue,the main mechanism is that WGPA promotes gluconeogenesis by enhancing the activity of G6 Pase and PEPCK.For G6 Pase,WGPA promotes transcription of its mRNA to increase the amount of enzyme to promote its activity.But the regulation of PEPCK activity is still need further study.
