Objective : To explore effects of different exercise intensity combined with Resveratrol on blood glucose and insulin resistance,mRNA expression and protein expression on adiponectin in visceral adipose tissue,plasma adiponectin concentration of elderly obese rats Methods : The male SD rats were used to establish the elderly obese rats model.Natural growth of rats as the blank control group(n = 6),36 obese rats were selected and were randomly divided into 6 groups(n = 6),including obese control group,resting+Res group,low intensity exercise group,low intensity+Res group,moderate intensity exercise group,moderate intensity+Res group.The running speed of the exercise groups were low intensity 12m/min,moderate intensity 15m/min,15mins/group,4 groups/times,5 mins rest between groups,60min/ days,running a slope of 0°,5 times / week.Using 52.5mg/(kg?d)of Res to fill the stomach,5 times/w,8 weeks of continuous intervention.8 weeks after the acquisition of rat blood and visceral adipose tissue,QRT-PCR method was used to measure mRNA expression on adiponectin visceral adipose tissue and Western blot method to measure the protein expression of adiponectin visceral adipose tissue,ELISA method to measure the plasma insulin and plasma adiponectin concentration and insulin resistance was calculated.Results:1.m RNA expression of adiponectin in visceral adipose tissue: Obesity group was lower than control group but no difference(P> 0.05);Exercise,different intensity of exercise combined with resveratrol could significantly improve the expression of adiponectin,the amount of exercise intensity,moderate intensity exercise was significantly higher than low intensity exercise(P < 0.05,P < 0.01);The effect of adiponectin on resveratrol was small(P>0.05).2.Protein expression of adiponectin in visceral adipose tissue: Obesity group was lower than control group;Exercise,different intensity of exercise combined with resveratrol could significantly improve the expression of adiponectin,moderateintensity exercise was significantly higher than low intensity exercise;The effect of adiponectin on resveratrol was small.3.Plasma adiponectin concentration: Obesity group was lower than control group(P<0.05);Exercise,different intensity of exercise combined with resveratrol could significantly improve the expression of adiponectin(P < 0.05,P < 0.01),the amount of exercise intensity,affected no significantly by the intensity of exercise(P>0.05);The effect of adiponectin on resveratrol was small(P>0.05).4.Blood glucose and insulin resistance: Obesity group was higher than control group(P < 0.01);Exercise could significantly reduce insulin resistance,moderate intensity exercise was significantly higher than low intensity exercise(P<0.05,P<0.01);The different intensity of exercise combined with resveratrol could significantly reduce blood glucose and insulin resistance(P < 0.05,P < 0.01),affected no significantly by the intensity of exercise(P>0.05);The effect of blood glucose and insulin resistance on resveratrol was small(P>0.05).Conclusion: Through this experimental study:(1)The mRNA expression and protein expression of adiponectin in visceral adipose tissue and plasma adiponectin concentrations were significantly lower in the elderly obese rats,the exercise could increase the level of adiponectin;With the increase of exercise intensity,the increase was more obvious;Different intensity of exercise combined with resveratrol could improve the level of adiponectin,which was less affected by the intensity of exercise.(2)The blood glucose and insulin resistance were significantly increased in the aged obese rats.Exercise,different intensity of exercise combined with resveratrol could significantly reduce blood glucose and insulin resistance,and were less affected by exercise intensity.(3)Resveratrol can improve the gene expression,protein expression and plasma concentration of adiponectin,also can reduce the blood glucose and insulin resistance in obese rats with the stimulation of exercise.
