Objective: To investigate the genetic polymorphisms of 23 short tandem repeat(STR)loci in Zunyi Han population.The values of STR loci were evaluated in forensic to provide a calculative database for individual identification and paternity testing.We not only observed new mutations in the new site of the Huaxia PlatinumTMKit compared to the Identifiler? Plus kit but also analysis the rare alleles in the Zunyi Han population and compare them with other people.Methods: Collect statistical information on 609 unrelated healthy individuals in the case of Judicial Expertise Center,the Affiliated Hospital of Zunyi Medical College from 2016 to2017.Peripheral blood was collected during the case test and multiplex PCR amplification was performed using the Huaxia PlatinumTMKit and direct amplification method.The amplified product was subjected to capillary electrophoresis using the3500 DX Genetic Analyzer,and the result of electrophoresis was analyzed by Gene Mapper? ID software.Application of Power Stats v1.2 Software to Statistics and Calculation of Forensic Genetic Parameters of 23 Autosomal STR Loci.Observe and analyze mutations and rare alleles in the population and confirm the experiment with Identifiler? Plus or Goldeneye 20 A kits.Results:(1)The 23 autosomal STR loci from 609 unrelated individuals were calculated and the allele frequencies ranged from 0.001 to 0.524.All 23 STR locus allele frequencies were normally distributed.(2)The frequency of allele frequencies of the 23 autosomal STR loci was consistent with the Hardy-Weinberg equilibrium in the Zunyi Han population(P > 0.05).Among them,the D5S818 locus reached the Hardy-Weinberg equilibrium after Bonferroni correction.(3)A total of 272 alleles and 1022 genotypes were detected in the 23 autosomal STR loci in 609 unrelated individuals.The MP valuesranged from 0.014 to 0.212,the PD values ranged from 0.788 to 0.986,and the PIC ranged from 0.550 to 0.910.The PE values ranged from 0.325 to 0.822,the Ho values ranged from 0.627 to 0.913,the TPI ranged from 1.34 to 5.75,the CPD was1-3.77696×10-28,and the CPE was 0.99999999731265,indicating that the detected STR locus with high polymorphism.Strong ability to identify.(4)Compared with the Han ethnic group in Inner Mongolia,the lowest number of alleles in the 23 autosomal STR loci in the Han population in Zunyi area is TH01,but the Inner Mongolia Han population also included TPOX,while the Zunyi Han population had two additionalloci: D3S1358 and D16S5392.(5)The frequencies of the 23 alleles of the STR loci in the Han population in Inner Mongolia were between 0.001 and 0.521.26 alleles at 14 loci were not detected in this study,but were distributed among the Han population in Inner Mongolia,34 alleles at17 loci were not detected in the Inner Mongolia Hans,while the Zunyi Hans were distributed,but the normal distribution of the gene frequencies was similar.(6)In the 159 cases of Trios and 328 cases of Duos involved in this study,18 cases mutations were found to have been detected by two kits.One case of TPOX(8,9,11),one case of three-step mutation D13S317(8?11),and one case of off-ladder allele were found in Zunyi Han population,D21S11(OL = 30.3)for the first time.Conclusion:(1)The 23 autosomal STR loci included in the Huaxia Platinum TM Kit are highly genetically polymorphic in the Zunyi Han population.The allele frequency distribution reaches the Hardy-Weinberg equilibrium and is suitable for individual identification and paternity testing in the Zunyi Han population.(2)The alleles and their frequency distribution of the above 23 STR loci in Zunyi area and Inner Mongolia Han population have high similarities,but there are also some differences.(3)The discovery of rare alleles and rare three mutations enriches the genetic information data of the Han population in Zunyi.
