Short tandem repeats(short tandem repeat,STR) in the human genome are widely distributed,multi-located in a non-coding region.STR in groups have greater variation,but the family was able to stabilize the genetic,so STR for individual identification and paternity test provides a reliable and rich source of information,has been widely used in forensic science,organ transplants and other medical fields. Triplet detection technology application in paternity testing has become quite mature, but the lack of parental genetic information,little is currently carrying out identification of sibling relationships,these reported in the literature are relatively small.In this paper,STR polymorphisms of good sites and more stable genetic characteristics of co-dominant to autosomal STR-based testing,while detection of sex chromosome STR,tracing individuals from four pairs of sibling relationships identified,and to explore the short tandem repeat sequences in the relationship between the compatriots on paternity identification of the application of value and sources of STR alleles without parents,all the same / the whole number of different loci in the identification of the joint application of the compatriots on the feasibility of, in order to establish simple and reliable method of identification of sibling relationships to provide information. Methords:Require identification of known fellow compatriots(respectively 2,4,3,3 people), and determined individual(4) blood samples by SDS-protease - phenol - chloroform extraction method to extract genomic DNA,PCR multiplex amplification 24 one autosomal STR loci,four X-STR loci,as well as seven Y-STR loci,polyacrylamide gel electrophoresis and silver staining band,through the known genotypes of the compatriots on speculation his parents and other bits of the 4 genetic screening does not match the individual to be determined loci and to compare the known compatriots, and determined the matching of alleles between individuals,statistics compatriots and to be determined in individual controls all different,all the same number of loci to three More than loci without parental sources,or all the different loci in the number of≥9,or all the same locus≤1 sibling relationships as a basis for exclusion,to be determined to distinguish individuals.Results:Fellow groupⅡ,Ⅲ,Ⅳto be determined in the individual and the parents does not match the number of gene loci,respectively 9,6,8,can rule out the genetic relationship;fellow groupⅠin the two known compatriots,only 5 bits points inferred parental gene,of which two parents in the detection of STR loci alleles not exist,can not be excluded sibling relationships.4 determined the individual and the corresponding group known compatriots, compatriots in pairwise comparison,fellow groupⅢ,Ⅳto be determined in each individual with a known number of compatriots in all the same locus and the whole range of different criteria for judging the number of loci of the outside(both 4 and 8), sibling relationships can not be excluded,the rest are in line with all the same locus≤1 or total number of≥9 different loci of the conditions can be ruled out sibling relationships.Use of STR analysis and STR loci allele all the same / the whole method of combining different numbers of four individuals can make a tracing of the pending rule out the genetic relationship of the expert conclusion of compatriots. Conclusions:1,fellow identification of known compatriots,the number of≥3,the availability of more reliable genetic information.2,the application STR alleles to exclude non-parental sources of the same law and the whole / whole different loci in the number of calculations can be simple,rapid and accurate identification of compatriots to improve identification of compatriots of exclusion.
