Leukemia is a disease,which is because of the blood cells,mainly white blood cells have a series of abnormal tumor cell proliferation in all the tissues and organs lead to anemia,hemorrhage,infection and infiltration and other symptoms.In china,Acute leukemia has a higher incidence than chronic leukemia(about5.5:1),AML(acute myelocytic leukemia)with the highest incidence(1.85/10 0000)followed by ALL(acute lymphocytic leukemia)(0.69/100000),CML(chronic myelogenous leukemia)(0.36/10),CLL(chronic lymphocytic leukemia)is infrequent.But different types of leukemia patients and treatment effect differences,leukemia patients genome research will explore the possibility of personalized gene therapy and prevention.Human leukocyte antigen(HLA)not only controls the same graft rejection reaction,be closely related to the body's immune response,immune regulating and the generation of pathology,has been found that a large number of diseases associated with a particular HLA,a specific type of HLA become the genetic marker for some certain diseases,and the relevance between leukemia and HLA genotype were reported at home and abroad.A large number of studies have been shown that different HLA genotype has a difference influence on leukemia,the people from different regional have a different HLA genotype and their frequency are different,and different susceptibility to leukemia.The existing researchs on the correlation between HLA and leukemia results are not the same.This study collected the samples of three common leukemias:AML ?ALL ?CML,and some samples from the healthy unrelated people to get the HLA genotyping at 4-digit allelic level for HLA-A,-B,-C,-DRB1 and DQB1 loci.and analysis the results of the healthy unrelated people and the patients with different types.to get the relevance between the HLA genotypes and the incidence of leukemia.Methods:A total of 854 leukemia patients,including 269 patients with acute lymphocytic leukemia(ALL),415 with acute myelocytic leukemia(AML),170 with chronic myelogenous leukemia(CML),and1496 healthy unrelated people were typed,the Next generation sequencing(NGS)based,and supplemented by Sanger-sequencing to get the high resolution of HLA-A,-B,-C,-DRB1 and DQB1 loci.Conclusion: In CML group A* 02:03,C*07:02 genes frequency are significantly higher than the control group,and the difference is statistically significant(P < 0.01)corresponding relative risk ratio(RR 2.511,95% CI 1.615,3.902;RR 1.824,95% CI 1.394,2.384,respectively)in AML group A* 02:03,DQB1*05:02 genes frequency are significantly higher than the control group,the difference is statistically significant(P < 0.01)corresponding relative risk ratio(RR 1.933,95% CI 1.369,2.727;RR 2.031,95% CI 1.557,2.649,respectively)in ALL groups A* 02:03,B* 13:01,DRB1* 16:02,DQB1*05:02 genes are frequency significantly higher than the control group,the corresponding relative risk value of the difference is statistically significant(P < 0.01),(RR 2.323,95% CI 1.59,3.392;RR 2.147 95% CI 1.540,2.992;RR 2.29,95% CI 1.513,2.29;RR 2.239,95% CI 2.239 to 1.656,respectively)The above results showed that multiple alleles of HLA-A,HLA-B HLA-C,HLA-DRB1,HLA-DQB1 gene loci,in AML,CML,ALL three types of leukemia showed high risk allele or low risk allele,providing important clues for the clinical diagnosis of Chinese leukemia.
