Abstract/Summary:
Cortex Moutan is the dried bark of peony which belong to Ranunculaceae plants, its smell is bitter and slightly cold. Cortex Moutan got more and more widely study and attention is an important traditional Chinese medicinal materials, having extensive exploitation and application prospect. The result of Modern pharmacology indicated that Cortex Moutan has many varies functions, including anti-microbico and antivirus activity, hypoglycemic activity and anti-inflammatory action. It can effects many physiological processes as immune system, cardiovascular system and blood system. Recently, many research about chemical component analysis of Cortex Moutan have been reported. The main chemical components of Cortex Moutan are paeonol and its ramification, paeoniflorin and its ramification. The primary analysis methods are gas chromatographic method,HPLC(High Performance Liquid Chromatography) and thin layer chromatography.For this reason, this study will based on extraction efficiency of paeonol, screening out the best condition of Steam distillation method and Supercritical CO2 extraction method. Using the thin layer chromatography and ultraviolet spectrophotometry to compare Steam distillation method and Supercritical CO2 extraction method to the impact on paeonol. At the same time, the pharmacologic action of paronol in anti-microbico and anti-inflammatory has been preliminary studied, the detail resut are as follows:In the optimization of Steam distillation method, using Orthogonal test to investigate the Warm immersion temperature, soak time, the ratio of Cortex Moutan and liquid and salt ratio to the impact of extraction ratio of paeonol based on single factor experiment, the results show that the best extraction condition is:Warm immersion temperature at 40℃, soak time for 3h, the ratio of Cortex Moutan and liquid is 1:6 and the percent of solt is 1%.In the optimization of Supercritical CO2 extraction method, using Orthogonal test to investigate extraction temperature, extraction pression, the rate of flow of CO2 and separation temperature to the impact of extraction ratio of paeonol based on single factor experiment, the results indicate that the best extraction condition is:extraction temperature at 50 ℃, extraction pression at 23Mpa, he rate of flow of CO2 is 16L/h and separation temperature at 40 ℃. According to Orthogonal test data, the extraction ratio of paeonol as response value, the independent variables are extraction temperature, extraction pression and the rate of flow of CO2, building response surface equarion:Y=1.91-0.083 A+0.13B+0.100C+0.035AB+0.050AC-0.010BC-0.039A2-0.36B2-0.10C2 R2=0.9537Adopting thin layer chromatography to compare the standard substance of paeonol with extractive, results indicated that extractive and the standard substance of paeonol have the same spots on the thin layer plate. In another word, this extraction method can isolate paeonol from Cortex Moutan efficiently.The purified extractive and standard substance were scanned by ultraviolet spectrum, the result indicated that the purified extractive has the same component with standard substance and has a high extraction ratio.The result of HNMR(H nuclear magnetic resonance) showed that the structure of paeonol has five kind of hydrogen atom, which cooperate with the reported of pharmacopeia and literature, so Supercritical CO2 extraction method do not break the structure of paeonol.At last, we studied the paeonol interacted with familiar bacteria and fungal by AGAR plate method of drilling hole. The result indicated that paeonol has some extent repression to bacteria and fungal, while it has stronger repression to bacteria than fungal. Between E.coli and bacillus subtilis, paeonol has stronger repression to bacillus subtilis when the concentration of paeonol is just 0.2%. Between Aspergillus niger and trichoderma, Aspergillus niger showed stronger repression.Through the study of paeonol interaction with carrageenan which can result in swell of mice’s vola pedis, to clarify the anti-inflammatory of paeonol in vivo. The result indicated that paeonol can prevented mice’s vola pedis to swell obviously.
