This dissertation is composed of two parts:literature review and experimental research.The part of literature review:The first literature review:This paper reviews the recent research papers on psoriasis models.This paper is divided into four parts,the standard of psoriasis models,animal models,cell models and three-dimensional skin model,and it also summarizes the operation steps of these models and recent researches which use thoese models.This paper aims to provide the base of using the pathological models of psoriasis in future.The second literature review two:This paper reviews recent research papers on usingchinese medicine monomer to treat psoriasis,including Paeonol,Paeoniflorin,Shikonin,Paeony glycosides,Glycyrrhizic acid,Tripterygium glycosides and other monomers.This paper also summarizes these monomers' pharmacologic action and the progress of using monomers to treat psoriasis.This paper indicates that the traditional Chinese medicine monomer has broad prospects in treating psoriasis.OBJECTIVE:To observe the effect of paeoniflorin and paeonol on the expression of SOCS1,Stat3,TNF-?,IL-1?,IL-6 and INF-? in IMQ-induced psoriasis mouse model.METHODS:BALB/c mice were applied a 22 mg/cm 2 5%imiquimod cream once a day for 8 days to prepare an imiquimod-induced psoriasis mouse model after being shaved on the back.Meanwhile,we fed the mice 0.2 mL of the corresponding drug(Paeoniflorin low dose group 23.245mg/kg/d,paeoniflorin middle dose group 46.49mg/kg/d,paeoniflorin high dose group 92.98mg/kg/d,paeonol low dose group 1.175mg/kg/Paeonol middle dose group 2.35mg/kg/d,paeonol high dose group 4.7mg/kg/d).We used the macroscopic evaluation and PASI to compare the changes of the lesion in each group.This experiment used immunohistochemistry to detect the expression of SOCS1,Stat3 and JAk1 in the lesions of each group,used PCR,Weestern Bolt to detect the expression of SOCS1,Stat3 and JAk1 in each group.Results:(1)There were significant differences in the expression levels of SOCS1,Stat3,TNF-?,IL-1?,IL-6 and INF-y in the model group(P<0.05),HE staining and visual observation,PASI score.Indicating that the model group than the blank group of psoriasis-like lesions heavier,suggesting that modeling success.(2)The expression levels of SOCS1,Stat3,TNF-?,IL-1?,IL-6 and INF-y in triptolide and paeoniflorin and paeoniflorin were significantly higher than those in model group(P<0.05),HE staining and visual observation,PASI score showed that the model group than the drug treatment group lesions and pathological manifestations lighter than the model group,suggesting that drug treatment is effective.(P<0.05).The levels of TNF-?,IL-1?,IL-6 and INF in the high dose group were significantly lower than those in the model group(P<0.05)(P<0.05).(P<0.05).Conclusion:There is a significant difference between the two groups(P<0.05),but there is no significant difference between the low dose group and the low dose of paeoniflorin(P<0.05).Conclusion:JAK1/Stat pathway is highly expressed in the lesion of psoriasis mouse model,and SOCS protein level is positively correlated with JAK1/Stat pathway activity.Paeoniflorin can inhibit the expression of downstream inflammatory factors TNF-?,IL-1?,IL-6 and INF-y by inhibiting the activation of JAK1/Stat pathway in psoriasis mouse model by regulating SOCS1 protein level,and In the lower dose group,paeoniflorin in the high dose group,paeonol in the high dose of therapeutic effect is stronger.
