Objective:To study the curative effect of curcuma zedoary on primary liver cancer,to compare with the difference of curative effect between raw and vinegar zedoary.To further explore the possible mechanism of acting on primary liver cancer with zedoary.Methods&ResultsThe experiment mainly consists of two parts.1.In vivo experimental study of raw and vinegar zedoary.SD rats were injected with 50%CCl4 neutral rapeseed oil solution in the double hind limbs alternately after one week of adapting feeding.After 5 weeks,DEN solution was prepared by sterile tap water(protected from light and freshly prepared),rats were free to drink,continuous drinking for 90 days,establishing the liver cancer models,and taking part of the liver tissue for pathological examination to determine whether the success of modeling.At the 17th week,the rats were given gavage orally.After 42 days of continuous administration,the abdominal aorta of each group was taken blood and the liver tissues were harvested.Serum levels of AFP,AFP-L3,IL-17 were detected by ELISA.The results were that the levels of AFP,AFP-L3 and IL-17 of serum in the groups of raw and acetic zedoary were lower than those in the group of models and higher than those in the group of blank control and the group of positive drug(p<0.05);The levels of AFP,AFP-L3 and IL-17 of serum were higher in the group of raw high zedoary than those in the group of acetic high zedoary,serum levels of AFP,AFP-L3 and IL-17 were higher in the group of raw low zedoary than those in the group of acetic low zedoary(p<0.05);the levels of AFP,AFP-L3 and IL-17 of serum in group of the raw high zedoary were significantly lower than those in the group of raw low zedoary,while those in the group of acetic high zedoary were lower than those in the group of acetic low zedoary(p<0.05).2.In vitro experimental study of the medicated serum about raw and vinegar zedoary acting on human liver cancer cells named HepG2.a.We prepared the drug-containing serums.It was divided into the blank drug-containing serum group,the drug-containing serum of raw zedoary,the drug-containing serum of vinegar zedoary,the positive drug-containing serum group.b.Using CCK-8 experiment to observating the growth inhibition of HepG2 cells by putting in medicated serum about different concentrations of raw and vinegar zedoary(5%,10%,20%,40%),setting the blank drug-containing serum group and the positive drug-containing serum group(20%drug-containing serum of cyclophosphamide group),acting with 24h,48h and 72h respectively.The results were listed on the table,and the optimum serum concentration of zedoary acting on cells was obtained.c.According to the experiment of Western-blot to observating the effect of medicated serums of raw and vinegar zedoary(20%)on AKT protein and p-AKT protein in HepG2 cells.Setting the blank drug-containing serum group and the positive drug-containing serum group(20%drug-containing serum of cyclophosphamide group).The drug-containing serum acting on the cell for 48h,testing the level of AKT protein and p-AKT protein in HepG2 cells by western-blotting.The results showed that compared with the blank drug-containing serum group,after the treatment with drug-containing serum about raw,vinegar zedoary and positive drug,there was no significant change in AKT protein of the drug-containing serum groups of the raw and the vinegar zedoary,the levels of AKT protein of positive control group were significantly decreased;the levels of p-AKT protein of the drug-containing serum of the raw,the vinegar zedoary groups and the positive drug group were decreased significantly,the decline levels were:the drug-containing serum of the raw zedoary group<the vinegar zedoary group<the positive drug group.Conclusion:The models of primary liver cancer in SD rats were successfully established.The results showed that the curative effect of raw and vinegar zedoary on rat liver cancer models were effective,but having less efficacy than the the po'sitive drug;vinegar zedoary has higher efficacy than raw zedoary,high concentration was better than low concentration.Zedoary has the effect of inhibiting the proliferation of tumor cells.And its inhibitory effect may be achieved by decreasing the levels of AKT phosphorylation through PI3K/AKT signaling pathway.
