| Objective：Ovarian cancer in female reproductive system is one of themost common malignancy with the highest death rate. There are not signalshows in the early phase of ovarian cancer and lack of early diagnosis methodso that most patients are always diagnosed in a later stage, even late widelytransferred,surgery has not completely removed. At present, the standardtreatment for ovarian cancer is cytoreduction surgery combined withchemotherapy. Cisplatin and other platinum-based cancer drugs are animportant class of the most active chemotherapeutic agents that are widelyused to ovarian cancer. However, acquired cisplatin resistance is a majortherapeutic problem for cancer chemotherapy. But in the course of treatmentabout80%of patients showed different degrees of acquired drug resistance,seriously affecting the prognosis of patients. Therefore, how to take advantageof new antitumor activity of the drug sensitive design best, with minimaltoxicity and drug resistance in chemotherapy scheme, is the focus of ourin-depth study,so as to improve the survival rate and quality of life of patientswith ovarian cancer.Zedoary turmeric oil is a kind of volatile oil monomer extracted from dryroots of Curcuma Longa plants. Zedoary is rich in volatile oil, the compositionis complicated, which is composed of β-elemene, Curcumenol, Curgerenone,Neocurdione, Camphor and so on.Zedoary turmeric oil is showed a lot ofeffects,such as anti-tumor,anti-virus,anti-inflammatory,anti-blood clot,immuneadjustment and so on，Especially,the antitumor function is widely concernedabout.Some researchs have turned out that the Zedoary oil cananti-proliferation of gastric cancer cells, liver cancer cells, cervical cancercells, endometrial cancer cells, etc. But the effect of Zedoary turmeric oil on cisplatin-resistant ovary cancer cell has not been reported.Studies suggest thatSTAT3signaling pathway and regulation of the downstream target genes canbe used as the mark of tumor chemotherapy drug resistance.To investigate the inhibitory effects of Zedoary Oil in the epithelialovarian cancer cell lines SKOV3and the cisplatin-resistant ovary cancer celllines SKOV3/DDP,and to discuss the expression of the STAT3pathway itspossible mechanism. Method: The inhibition and cellular morphology bydifferent concentration of Zedoary Oil in the proliferation of SKOV3andSKOV3/DDP cells was observed by light microscopy,HE staining. TheSKOV3and SKOV3/DDP cells were treated by different concentrations ofZedoary oil. rates of proliferation inhibition were detected by the means ofMTT.The SKOV3/DDP cells were treated with Zedoary Oil, cisplatin,Zedoary Oil together with cisplatin for24hours, to detect the proteinexpression of STAT3，Survivin, Caspase-3by immunohistochemistry andRT-PCR. Result: the number of SKOV3/DDP cells is significantlyreduced,while cellular morphology also obviously changed by lightmicroscope.the proliferation of SKOV3and SKOV3/DDP cells was markedlyinhibited by Zedoary Oil in a dose-dependent and time-dependent manner.(P<0.05). the protein expression of Caspase-3was activated.The expressionallevels of STAT3,Survivin proteins was decreased significantly byimmunhistochemistry and RT-PCR.Conclusion: Zedoary Oil can inhibit theproliferation of human epithelial ovarian cancer cell lines SKOV3andcisplatin-resistant ovary cancer SKOV3/DDP cell line in the study of vitro cellculture, and induce apoptosis. The down regulation of STAT3,Survivin,proteins expression may be related to cell growth inhibition and apoptosismediated by Zedoary Oil. The mechanism may be related to down-regulatingthe expression of Survivin, VEGFC,and to up-regulating the expression ofCaspase-3protein. The result of Spearman correlation coefficient analysis theexpressional levels of Survivin,VEGFC is associated with the expression of STAT3in SKOV3/DDP cells (P <0.01)... |
