Background: epilepsy(EP)is a clinical multiple causes of brain neurons highly synchronized caused by abnormal discharge syndrome,cause the cause of epilepsy is very complex,including genetic factors,growth factors,perinatal injury,brain inflammation,cerebrovascular disease,brain tra uma,brain tumor,nerve degeneration etc..At present,the treatment of epilepsy is still mainly drug treatment.However,even if a reasonable dose of Antiepileptic drugs(AEDs)is taken,about 30% of the patients with epilepsy develop Intractable epilepsy(IE).IE is poor,with high disability and death rate,which seriously affects the quality of life and social function of the patients.Therefore,we have studied the causes and mechanisms of drug-resistant epilepsy,and found that the new therapeutic measures are far-reaching.Chinese medicine has become a new breakthrough in recent years to improve the cure rate of IE,reduce the adverse reaction and drug resistance due to its action mechanism different from western medicine.Epileptic seizure is closely related to oxidative stress,P-glycoprotein and brain injury.Combined with traditional Chinese medicine in the treatment of intractable epilepsy,this study explored the possible mechanism of Chinese medicine compound in the treatment of intractable epilepsy,and it is also a major trend in the research direction of epilepsy.Objective: To investigate the effect of Chaihu Shugan Tang combined with Guipi Tang in the treatment of intractable epilepsy(IE)of liver stagnation and spleen deficiency syndrome clinical curative effect and anti oxygen free radicals,P-glycoprotein(P-gp)and insulin-like growth factor-1(IGF-1)on the expression of.Methods: according to IE diagnosis standard of liver stagnation and spleen deficiency syndrome in 60 patients,were randomly divided into control group and experimental group with 30 cases in each group,the control group was given routine treatment,while the experimental group was given on the basis of routine treatment,given Chaihu Shugan Tang combined with Guipi Tang Treatment,continuous treatment after 3 months.Video Electroencephalogram(EEG),after treatment detection;recorded before and after treatment of epileptic seizure frequency of epilepsy patients,seizure duration,Quality of Life in Epilepsy-31(QOLIE-31)were detected before and after treatment;serum total superoxide dismutase(SOD),Malondialdehyde(MDA)),P-gp,IGF-1 levels before and after treatment;calculating the number of seizures,seizure duration,difference of the scores of QOLIE-31,SOD,MDA,P-gp and IGF-1 levels.Results: the frequency of seizures in experimental group after treatment was lower than that of control group(P<0.05),the duration was significantly lower than the control group(P<0.01),the number of epileptiform discharges was less than that of the control group(P<0.05),QOLIE-31 scores were significantly higher than the control group(P<0.01),the level of SOD was significantly higher than the control group(P<0.01),MDA and P-gp levels were significantly lower than that of the control group(P<0.01),IGF-1 level is lower than the the control group(P<0.05);and control group difference comparison,the experimental group obviously reduce the degree of seizure frequency(P<0.05),the duration of less obvious(P<0.01),QOLIE-31 score increased significantly(P<0.05),SOD increased and P-gp decreased significantly(P<0.01),the degree of MDA and IGF-1 decreased obviously(P<0.05),the difference was statistically significant.Conclusion: Chaihu Shugan Tang combined with Guipi Tang in the treatment of IE patients with liver stagnation and spleen deficiency can reduce the seizure frequency and the duration of seizures,epileptiform discharge,improve the quality of life of patients,the mechanism may be related to scavenging oxygen free radicals,improve antiepileptic drugs(AEDs)reduce the damage of the brain,less sensitivity.
