Ojective:On the basis of study on the effect of Yiqi Huoxue Recipe on intestinal micro ecological side effects of omeprazole in patients with peptic ulcer,by expanding the sample,increasing the type of syndromes,verify the preliminary experimental results through the study of peptic ulcer patients and healthy population differences in intestinal flora,similarity and related species composition,clarification the difference of intestinal microflora in different type of syndromes of peptic ulcer,through further analysis of differences between different type of syndromes and intestinal microflora,and combined with the related experiments of patients with peptic ulcer symptoms room checks from the intestinal flora angle for type of syndromes of peptic ulcer type and treatment of traditional Chinese medicine to provide the experimental basis and objective basis.The use of modern technology to improve the advantages of medical treatment of peptic ulcers in the motherland,and promote its application,for the research of the Chinese medicine to find a new way.Methods:Patients with peptic ulcers and healthy volunteers were used as research subjects.Patients with peptic ulcer were from the outpatient and inpatient department of the spleen and stomach department of Suzhou traditional Chinese medicine hospital.The establishment of Chinese and Western medicine diagnostic criteria,inclusion and exclusion criteria,peptic ulcer symptom classification of quantitative criteria,all the cases were evaluated according to the above standard,and by gastroscopy and pathological diagnosis,symptoms score and be taken for bacteria detection of fecal samples,healthy volunteers only collected fecal samples for bacteria detection.The faecal DNA was extracted and the microbial diversity in the intestinal flora 16SV3+V4 region was analyzed based on the MiSeq high throughput sequencing platform.Based on 16SV3+V4 bioinformatics data,combined with symptom grading,quantitative score and laboratory examination,we analyzed the similarities,differences and correlations between different type of syndromes of peptic ulcer and healthy population,as well as the structure of intestinal flora between different type of syndromes.Results:The biological information analysis platform according to Miseq sequencing:four peptic ulcer syndrome compared with healthy people,the difference of species richness was not statistically significant(P>0.05);patients with peptic ulcer can be healthy people between the species diversity of the difference was not statistically significant(P>0.05);the gate level,comparecd with healthy people,weak card type temper actinobacteria patients compared with healthy individuals(P<0.01.reduce the difference is highly significant),damp heat syndrome of spleen and stomach in patients with verrucomicrohia decreased compared with healthy people(P<0.01,the difference is highly statistically significant);compared between the four groups of syndromes of peptic ulcer,spleen deficiency and stomach found stasis syndrome actinobacteria is deficiency of Spleen Qi syndrome patients(P<0.05,the difference was statistically significant),verrucomicrobia and Cyanophyta than patients with spleen stomach damp heat syndrome(P values were more than Less than 0.05,the difference was statistically significant);patients with abnormal cocci-spleen stomach damp heat syndrome of Thermus door than the other three groups of syndromes in patients(P values were less than 0.01,the difference is highly significant);several other bacterial phyla although each have differences,but by statistical analysis,there were no significant;the species level,species differences have statistical significance between samples are the following:Brevundimonas?Curvibacter?Synergistes?Neisseria?Granulicatella?Weissella?Massilia?Atopobium?Streptococcus?Anaerotruncus?Bacillus?Sneathia?Hungatella?Peptoniphilus?Aggregatibacter?Desulfovibrio?Hydrogenoanaerobacterium?Fretibacterium?Corynebacterium?Slackia?Solobacterium?Oscillibacter?Dielma?Achromobacter?Parasutterella;the type of syndromes grading score,type of syndromes clinical analysis showed that patients with Qi deficiency of spleen and stomach There were significant differences in symptom scores compared with the other three syndromes,and there was no significant difference between the other three groups.Conclusions:The clinical symptoms of patients with spleen-and-qi deficiency syndrome are more serious than those of other three syndromes;there is no significant difference in intestinal flora diversity between patients with peptic ulcer and healthy people;In contrast,there was no significant difference in the diversity of the intestinal flora;there were differences in the level of some gates and species between the species composition of the gut microbiota of different type of syndromes of peptic ulcers.The level of type of syndromes on the level of the door can be distinguished from the actinomycetes to the deficiency of temperament.Type of syndromes classification on the level of the gene may be related to pathogenic bacteria and harmful bacteria.
