Background Metabolic Syndrome(MS)is widespread prevalence and obviously rising around the world.At present,the etiology and pathogenesis of MS is still far from very clear,and the treatment has no major breakthrough.In recent years,the relationship between the “imbalance intestinal microecological" and the occurrence and development of metabolic disease has become a hot research at domestic and overseas.Studies have shown that imbalance of intestinal flora play an important role in the occurrence and development of diabetes,obesity.As we know,MS has common soil with them— insulin resistance;In addition,many of researchs has confirmed that Spleen deficiency be closely related to imbalance of intestinal flora,and MS have the closest relationship with the "spleen".Therefore,We speculate that the occurrence and development of MS may be some correlations with intestinal flora.Currently,the study on intestinal flora and metabolic diseases mostly concentrated in obesity and type 2 diabetes in the domestic and foreign.The research of the relativity with MS only limited to the level of animal experiments,and so far research on human intestinal structure and quantity system have not been reported.MS as a "high risk" and "early stage" of variety major chronic diseases,it is of great significance to study in-depth.Research purpose 1)The research is aim to compare intestinal flora quantity differences,include nine class main flora of four euryarchaeotas,in MS patients and healthy of middle-aged and old people by real-time fluorescent quantitative PCR technology(Q-PCR).And then,to expound the correlation between intestinal bacterial flora and MS,to seek for new clues for the pathogenesis and new targets for prevention and control of MS.2)To explore the correlation of intestinal flora with syndrome of spleen deficiency phlegm-dampness of MS,which would provide theoretical basis for Syndrome differentiation type,reveal the essence of spleen deficient phlegm dampness syndromes and guide Traditional Chinese medicine treatment of MS.The study included 80 MS patients(40 phlegm-dampness due to spleen deficiency and 40 Qi and Yin deficiency syndrome),according to CDS(2013)MS diagnostic criteria,and 30 healthy volunteers.Fecal samples were collected from all subjects.Total DNA was extracted from collected stool samples and submitted to Q-PCR with primers specifically targeting V3 region of the 16 sr RNA gene,standard curve made by plasmid as standard substance.MethodResults 1)Comparison of the bacteria between MS and health control group There were significant differences between MS and healthy persons by comparing total bacterial,bifidobacterium,enterococcus,lactobacillus,clostridium cocc oides,clostridium leptum,Escherichia coil.Total bacteria,bifidobacterium,lactobacillus,enterococcus in MS were signifiant fewer than the healthy control group(P < 0.05),clostridium coccoides,clostridium leptum,escherichia coli were signifiant higher than control group(P < 0.05).2)Comparison of the bacteria among three group(MS spleen deficient phlegm dampness syndromes,Qi and Yin deficiency syndromes and healthy controls).There is statistically signifiant difference in the total bacteria,clostridium coccoides,bifidobacterium,lactobacillus,clostridium leptum,enterococcus,escherichia coli among three groups(P<0.05),and further comparsion:(1)MS Spleen deficient Phlegm Dampness syndrome group were lower in the number of total bacteria,bifidobacterium and lactobacillus,higher in clostridium coccoides,clostridium leptum,escherichia coli compared with healthy controls,the difference is statistically significant(P<0.05).(2)Spleen deficient phlegm dampness syndrome compare with Qi and Yin deficiency syndrome of MS,the number of bifidobacterium significantly reduced,escherichia coli amounts increased significantly(P<0.05).(3)MS of Qi and Yin deficiency syndrome group compared with the healthy group of decrease in the number of total bacteria,increase in enterococcus with significant difference(P<0.05).Conclusion 1)The result of this study indicate that MS is associated with compositional changes in intestinal flora.2)Intestinal flora of MS exist differences between different TCM syndrome types.3)The imbalance of Intestinal flora plays an important role in spleen deficient phlegm dampness syndromes of MS,the change of intestinal bacteria can be used as an objective indexes of spleen deficiency.
