Antrodia camphorata is a kind of medicinal mushrooms,which contains a variety of pharmacological activity compounds,for instance terpenoid,polysaccharide,protein,benzoquinone,aromatic,lipids,etc.These active substances endowed antrodia with a very high medicinal value such as to protect liver,anti-inflammatory,anti-allergic,anti-virus,anti-oxidant,regulating immunity and tumor therapy.Melanocytes derive from neural crest cells,is the site of melanin synthesis and secretion.Pevious researches demonstrated that melanocyte also involved in skin immune reaction process.Multiple diseases could be brought out for the dysfunction of melanocyte,as albinism,vitiligo and so on.Experimental purposesIn this research,we investigate the impact of the commercial antrodia on melanocyte,especially cell proliferation,apoptosis,factor,the rate of melanin synthesis and cell migration.MethodsPrimary cell extraction: With reference to published method,melanocytes were got from the the children foreskin tissue by the enzyme digestion.Positive results got from L-dopa dyeing and immune verification demonstrate the high purity of melanocytes.Activity research: MTT was employed to test the influence of antrodia on the proliferation of melanocytes.And an apoptosis model induced by oxidative stress of L-dopa was employed to research the protective effects of antrodia.Function research: The level of proinflammatory cytokines was detected by Elisa kit.Tyrosinase activity in melanocytes was detected by reaction rate method of L-dopa.The amount and the rate of melanin synthesis were explored by cell lysis with NaOH.Finally,transwell method was employed to measure cell migration rate.Result:1.According to the results of L-dopa dyeing and immunosorbent validation,pure primary melanocytes were extracted successfully.Uniform cell morphology excluding keratinocytes and interconnected enation were obtained by microscope.2.In accordance with MTT,melanocytes proliferation was obviously facilitated with antrodia existing.Simultaneously,antrodia could protect melanocytes from apoptosis induced by oxidative stress.3.In the function research,a few favorable phenomenons were obtained such as reducing of proinflammatory cytokine,activation of tyrosinase activity,enhancing of melanin synthesis and promotion of the melanocyte migration.Conclusion:In this research,human primary melanocytes were extracted successfully.According to the results,antrodia showed great pharmaceutical potentials,protecting melanocyte from apotosis,reducing proinflammatory cytokine secretion,enhancing melanin synthesis and promoting melanocyte migration,which were promising to be applied in vitiligo therapy.
