Objective:?1?To observe Pu Jin oral liquid on the pathological change and the expression of nogo receptor?NgR?and neurotrophin receptor?p75NTR?in the brain tissue of premature rats with intrauterine infection/inflammation caused by intrauterine infection induced by LPS.?2?To compare the effect of high and low dose Pu Jin oral liquid,Ginkgo biloba extract and early intervention and late intervention on nerve myelin's regeneration and repairment in rats with premature damaged brain.?3?To explore that"phlegm"and"stasis"may be pathological factors for premature infants with brain damaged caused by intrauterine infection/inflammation and provide experimental evidence for early clinical intervention of Chinese herbs for promoting blood circulation to remove blood stasis and resolving phlegm.Methods:?1?Pregnant rats were randomly divided into lipopolysaccharide?LPS?group?n=30?and control group?n=6?according to the random number table method.Intraperitoneal injected the pregnant rats with LPS at a dose of 350?g/kg or isodose saline on the 16th and 17th days of pregnancy.Defined premature rats according the Gestational age which less than or equal to 21days.?2?Divided 80 premature rats into 8 groups?n=10?randomly of which A1 stands for the early intervention group of high dose of Pu Jin oral liquid,A2 stands for the late intervention group of high dose of Pu Jin oral liquid,B1 refers to the early intervention group of low dose of Pu Jin oral liquid,and B2 refers to the late intervention group of low dose of Pu Jin oral liquid,C1is the early intervention group of Ginkgo biloba extract,and C2 is the late intervention group of Ginkgo biloba extract,D1 stands for the early intervention model group and D2 stands for the late intervention model group.?3?The early intervention group received gastric perfusion on the first day,while the late intervention group began on the eighth day.After the intervention all rats were fed to 21-days-old under the same condition.?4?Observe the pregnant and delivery situation of female rats and the birth weight,color,reaction ability and activity about newborn rats between the LPS group and the control group.Six of the pregnant female rats and 10 newborn premature rats were choosed in each groups randomly,and HE staining was used to observe the pathological changes of the uterus and placenta of the pregnant rats and the brain tissue of premature rats.Detected the expression of NgR and p75NTR protein in the brain tissue of the the pregnant PVL rats by immunohistochemistry and Western Blot on 0-day-old and 21-days-old.Results:?1?Compared with the 1-day-old rats of control group,LPS group has high abortion rate and less number of delivery?P<0.05?,and it is easy to occur premature delivery even stillbirth in general.Compared with control group,the LPS group's newborn rats has less activity,darker skin color,weaker stress and lower body weight?P<0.05?.?2?HE staining results about LPS group'uterine and placental tissue show a large number of inflammatory cell infiltration,while the control group'histopathological examination shows no inflammatory reaction;LPS group of preterm birth pathogens pathological examination present that tissues'structure of white matter district are sparse and wide,and have an irregular arrangement of nerve cells,while the tissue structure of the control group is complete,the cells are arranged neatly and the nuclei are larger.?3?Compared with 0-day-old rats in control groups,The expression of NgR and p75NTR protein in brain tissue of of LPS group increased?P<0.05?.In the early intervention group,the expression levels of NgR and p75NTR protein in the brain tissue of A1?B1 and C1 groups are significantly lower than those in model group?P<0.05?.The effect of high dose of Pu Jin Oral Liquid on the expression of NgR and p75NTR is significantly better than that of low dose Pu Jin Oral Liquid and Ginkgo biloba extract?P<0.05?.there is a significant difference in the expression of NgR and p75NTR in the low dose of Puji oral solution and Ginkgo biloba extract,and Ginkgo biloba extract is bettter?P<0.05?.In the late intervention groups,the expression of NgR and p75NTR in the brain tissue of A2?B2 and C2 groups are significantly decreased compared with D2?P<0.05?.But the effect of high dose of Puji Oral Liquid on the expression of NgR and p75NTR is significantly better than that of low dose Pujin oral solution and Ginkgo biloba extract?P<0.05?.Ginkgo biloba extract has obvious effect in down-regulating the expression of NgR and p75NTR compared with the low dose of Pu Jin oral liquid?P<0.05?.Compared with the late intervention group,the early intervention group with the same kind of drug are lower on expression of NgR and p75NTR,and the difference is statistically significant?P<0.05?.Conclusion:?1?Intrauterine infection/inflammation can upregulate the levels of myelin-associated inhibitory factor receptor of NgR and p75NTR in disabled brain tissue of premature infants with PVL.?2?Pujin Oral Liquid could inhibit the expression of NgR and p75NTR in brain tissue of premature rats,and the effect of early drug intervention was more significant.?3?The study conjectured that the pathologic factors of premature infant with brain injury caused by intrauterine infection/inflammation may be"phlegm"and"blood stasis".The mechanism of Chinese herbs of promoting blood circulation to remove blood stasis and resolving phlegm in treating PVL in premature infants may be inhibit the expression of myelin-related inhibitory receptor and promote the regeneration and recovery of nerve function.
