Objective: To observe Nogo-A, Ng-R mRNA expression in the damaged neonatal rat brain after intrauterine infection.Method: 1. Establishment of model: 40 Wistar pregnant rats were randomly divided into two groups: LPS-treated group (n=20) and saline control group (n=20). Pregnant rats were consecutively injected with LPS (450μg/kg) or saline on gestation d 17 and 18. The pups whose gestation age was less than 22 days were preterm pups. The interval from injection to delivery of each group was recorded, along with the number of live born pups and their birth weight. The preterm pups were removed in both groups. After delivering rat, a part of female rats were executed. The uterus and placenta were taken out. The H&E stained sections were examined under microscope for numbers of neutrophilic granulocyte in the uterus and placenta. It was evidence of infection that the numbers of neutrophilic granulocyte increased. Respectively selected randomly 40 rats from LPS neonatal rat group and saline neonatal rat group, divided them into 4 groups: 6h group (n=10), 12h group (n=10), 24h group (n=10), 48h group (n=10), punctually, the rats were executed.2. By using Real Time RCR (RT-PCR) method the mRNA expression levels of Nogo-A and NgR were determined.Results: 1. 4 pregnant rats were dead after injected LPS. There were4 pregnant rats of pre-parturition in LPS group. The rest of pregnant rats delivered 74 live mature rats and 39 dead rats in LPS group. In reverse, there were no dead or pre-parturition's pregnant rats in saline control group. The pregnant rats delivered all 184 live mature rats and no dead rats in saline control group. Compared with the neonatal rat of control group, there were purple skin and low weight in neonatal rat of LPS group. There were a large amount of neutrophilic granulocyte in the uterus and placenta in LPS-treated group.2. Saline neonatal rat group did not alter at different time points. Compared with saline neonatal rat group, in LPS neonatal rat group, Nogo-A mRNA started to increasing at 6h and reached the peak at 12h; NgR mRNA started to increase at 6h and reached the peak at 12h and kept increasing till 24h.Conclusion: 1. Compared with saline neonatal rat group, in LPS neonatal rat group, Nogo-A mRNA started to increasing at 6h and reached the peak at 12h; 2.NgR mRNA started to increase at 6h and reached the peak at 12h and kept increasing till 24h;3. The Nogo-A and NgR may have certain relation with obstructing the rebirth of the nerve which root in damaged neonatal rat brain after intrauterine infection..
