Objective: Observed the influence which resulted of the Danshen injection on theexpression of Nogo receptor and P75neurotrophin receptor which in the white matter ofbrain damage caused by intrauterine infection. Explore the mechanism of action of Dansheninjection to improve the intrauterine infection-induced brain damage in premature.Method:1. The30rats were divided into lipopolysaccharide (LPS) group (n=24), injectedLPS with350μg/kg·d on the first16th,17th days of pregnancy, the control group (n=6)were intraperitoneally injected at the same dose of saline.we called pups which birthed lessthan21days pregnancy as the premature rats. After the birth, we take out the uterus andplacenta and has the HE staining in order to observed the intrauterine infection。Observed thebirth weight color, activity, response capability of the two groups little rats。Take the braintissue of the LPS group and the control group, all the tissue has the HE staining to observethe periventricular white matter injury case.2. Selected60preterm rats randomly in the LPSgroup, they were divided into6groups of10rats. Such as,The early intervention group A1、B1、C1,late intervention group A2、B2、C2,and theA1、A2is the group of Salvia injection(9ml/kg·d), and the B1、B2is the group of Ganglioside group, the C1、C2is the NS group(9ml/kg·d). Randomly selected control group of10pups as a control group D.The earlyintervention group will have the intraperitoneal injection on the first day after birth, the lateintervention group will have the intraperitoneal injection on the eighth day after birth, theyall been injected for7days. After injected, all the rats adopted to the21th day at the samecondition, they will have the Unified interbehavioral tests(take the suspension test,the ramptest and the Cylinder test).3. we take the8groups rats brain tissue after they were been fedwith21days at the same conditions, we test the expression of NgR、p75NTR byimmunohistochemical method.Results:1. There are many neutrophil infiltration on the pregnant rat uterus and plac enta pathology and have the vascular congestion, edema after the Pathological examination in the LPS group. And there is no inflammatory response in the the control group. Compared with the control group, the LPS group have the lower weight, lessactivity, skin color cyanosis, the preterm rats in LPS group have the white matter internal capsule and the Corpus callose structure sparse and we can see the internal hemorrhage in the ventricle and white matter.2. Section21d neurobehavioral testing, after Danshen injection and ganglioside injection early intervention two pups neurobehavioral able less than the control group (P<0.05), improved than the model group(P<0.05), neurobehavioral capability performance between Danshen Injection and Ganglioside Injection are no difference (P>0.05);after Danshen injection and Ganglioside injectionand late intervention two groups pups neurobehavioral able less than the control group (P<0.05), improved than the model group (P<0.05), two pups neurobehavioral capability performance of Danshen injection and Ganglioside injection have no difference(P>0.05).3. After Danshen injection and Ganglioside injection early and late intervention expr ession of P75NTR and NgR in pups white matter are higher than the control group (P<0.05), lower than the model group (P<0.05), The role of Danshen injection down the expression of NgR and P75NTR in the pups white matter after early intervention is significantly higher than Ganglioside injection(P<0.05), the role of Danshen injection down the expression of NgR and P75NTR in the pups white matter after late intervention is group no difference with Ganglioside injection(P>0.05);4. Afterearly intervention the neurobehavioral capacity of Danshen injection and Ganglioside injection pups improved than after late intervention (P<0.05), the expression of NgR and P75NTR in pups white matter are lower than the late intervention expression.Conclusion:1. This study demonstrated that Danshen Injection has obvious improvement onintrauterine infection caused by PVL pups behavior.2. Danshen injection can reduce theexpress of NgR、 P75NTR which caused by intrauterine infection.3. The efficacymechanism induced by injection of intrauterine infection of peri-ventricular leukomalaciamay have the relation of the falling the express of NgR、P75NTR.
