Objective:?1?To observe the influence by pujin oral liquid on apoptosis of brain tissue pathology?neuroethology?oligodendrocytes?OL??oligodendrocyte precursor cells?OPC?of the preterm birth and brain injuried rats which caused by intrauterine infection or inflammation.?2?To compare the role on myelin regeneration and repair of the preterm birth and brain injuried rats caused by high and low dose pujin oral liquid?ginkgo biloba extract?early and late intervention.?3?To discuss and establish that phlegm and stasis is the main pathological basis of the brain injuried of the premature babies which caused by intrauterine infection or inflammation.It will provide experimental evidence for intervention treatment of the blood circulation phlegm resuscitation drug in the early.Methods:?1?Animal model making: 30 pregnant rats are randomly divided into lipopolysaccharide?LPS?group?n=24?and the control group?n=6?,They will be injected in intraperitoneal with LPS 350ug/kg and the same dose NS.At 16-17 days after pregnant for 2 days.Pups which confined in pregnancy < 22 days is the premature rat.?2?Experimental animal group: 80 Premature rats are randomly divided into 8 group,10 rats in every group.as follows:Early intervention group A1?B1?C1?D1,The late intervention group A2?B2?C2?D2,among them,A1?A2 are low dose pujin oral liquid group?B1?B2 are High dose Pu Jin Oral Liquid group,C1?C2 are ginkgo biloba extract group,D1?D2 are model group.?3?Intervention methods: Early intervention group are lavage administration on the day of birth,The late intervention group lavage administration 7 days after birth,two groups are all used in the continue 7 days.After the intervention,they all will be raised until 21 days after birth.then,neuroethology test?suspension test?slope test?cylinder test?.?4?Observation index: Maternal situation of the mother rats after modeling.Intrauterine infection orinflammation situation of the uterus and placenta by HE dyeing immediately after birth.Birth weight?color? ability of reaction?activity of the premature rats in the two groups.Detect the damage and apoptosis of the OL and OPC by immunofluorescence staining on the day of birth,which will be done once again 21-days-old on the premature rats in 8 groups.Results:?1?The general situation: the mother rats after modeling in LPS group are easier to Abortion and premature birth,and died in childbirth.The litter number in LPS group is less than the control group?P<0.05?.Compared with the control group,the weight of the premature rats in the LPS group is lower?P<0.05?.and also with less activity?purple purple skin color?weak stress response.?2?The pathological morphological changes after HE dyeing: There are congestion of blood vessel and a large number of neutrophil infiltration in the uterus and placenta tissue of the pregnant rats in the LPS group.But there is no inflammatory reaction in the control group;and there is sparse structure of brain white matter?internal capsule and the corpus callosum,also with malalignment of nerve fibers,Small number of neurons,bleeding and glial cell aggregation of ventricle and white matter.?3?Neuroethology capacity: Compared with the control group,High and low dose pujin oral liquid?ginkgo biloba extract all can improve the neuroethology capacity of the premature rats?P<0.05?.And the effect of early intervention is better than the late intervention?P<0.05?.But there is no significant differences in improving the neuroethology capacity of the premature rats with high and low dose pujin oral liquid?ginkgo biloba extract?P>0.05?.?4?The results of oligodendrocytes?OL?and oligodendrocyte precursor cells?OPC?: Compared with the control group,the number of the OL and OPC in premature rats'?on the day of birth?brain tissue is significantly decreased in the LPS group?P<0.05?.In the early intervention group,compared with the control group,High and low dose pujin oral liquid?ginkgo biloba extract all can significantly inhibite the apoptosis of the OL and OPC?P<0.05?.Comparison between the above three groups,high dose pujin oral liquid is better than others?P<0.05?,But there is no significant differences in low dose pujin oral liquid and ginkgo biloba extract.In the later intervention group,compared with the control group,high and low dose pujin oral liquid?ginkgo biloba extract all can significantly decrease the apoptosis of the OL and OPC?P<0.05?.Comparison between the above three groups,high dose pujin oral liquid is better than others?P<0.05?.Compared with the later intervention,there is a more obvious effect in decreasing the apoptosis of the OL and OPC in the early intervention group.Conclusion:?1?Intrauterine infection can lead to neuroethology changes of the premature rats and making the significantly decreasing of the number of the OL and OPC.?2?High dose pujin oral liquid can significantly inhibite the damage and apoptosis of the OL and OPC in premature rats' brain tissue caused by intrauterine infection,also,there is a more obvious effect with early intervention.It provides an experimental evidence for intervention treatment of the blood circulation phlegm resuscitation drug in the early.?3?Research shows that phlegm and stasis is the main pathological basis of the brain injuried of the premature babies which caused by intrauterine infection or inflammation.
