Study On The Influence Of IFN-? And AQP-5 Of SS Model Mice Treated By Zeng Ye Runzao Decoction

Objectives:Establishing an animal model of Sjogren's syndrome(SS)by Immune induction method.To dynamic observe the effects of Zengye Runzao decoction on IFN-?(IL-?)and AQP-5(AQP-5)in SS model mice,and clear its therapeutic target,to further explore the mechanism and provide the basis for Zengye Runzao decoction treating of SS,but also for its clinical treatment of Sjogren's syndrome provide a strong experimental basis.Method:Select BALB/c mice were killed by cervical dislocation,remove both sides of submaxillary gland,submandibular glands and separated tissue surface appendages,said quality,cut and crushed before adding normal saline in the ice bath homogenizer homogenate,and then centrifuged in a 4 ? low temperature centrifuge to discard the supernatant and retain the supernatant to obtain the autoantigenic protein.The antigen was diluted to 800[mu]g/ml with sterile PBS,and an equal volume of CFA was added to make the antigen to a 400[mu]g/ml oil-water mixed emulsion.60 rats were randomly divided into 6 groups according to a digital random table:blank group,model group(MXZ),low dose group(ZYZ ?),medium dose group(ZYZ?),high dose group(ZYZ ?),western medicine treatment group(XYZ).On day 0,MXZ,ZYZ ?,ZYZ ?,ZYZ ? and XYZ mice were injected with 0.05 ml of antigen on each side of the groin after alcohol disinfection,i.e.0.1 ml of antigen was injected into each mouse.On days 2 and 7,0.05 ml of the combination vaccine was administered intraperitoneally to MXZ.ZYZ ?,ZYZ ?,ZYZ ? and XYZ mice,resulting in SS-like submandibular glandular manifestations and symptoms.Three groups of traditional Chinese medicine group were given Zengye Runzao Tang granule infusion solution.The blank control group and the model group were treated with deionized water by intragastric administration.The western medicine group was treated with total glucosides of paeony.The expression of IFN-? in supernatant of submandibular gland was measured by Elisa method.SP immunohistochemical staining was used to detect the expression of AQP5.Results:Zengye Runzao Decoction can reduce the secretion of IFN-?in SS model mice.The results of pathological observation showed that Zengye Runzao Decoction could up-regulate the expression of AQP5 and decrease the infiltration of lymphocytes in mice.Day 14 The above procedure was repeated to enhance immunization.Conclusion:Zengye Runzao Decoction can reduce the content of IFN-Y in glandular tissue,inhibit immune and inflammatory reaction,combined with the pathological section,Zengye Runzao Decoction can repair the submandibular gland and the duct structure of the SS model mice,and remove the lymphocytes infiltrating and accumulating,then restore the physiological function of the gland,up-regulate the expression of AQP5,achieve the purpose of alleviating the disease.
