Study On The Influence Of TNF-α、IL-6、IgG Of SS Model Mice Treated By Zeng Ye Runzao Decoction

Objective:To explore the mechanism of Zeng Ye Runzao decoction treatingSS (Sjogren’s syndrome).By studying the influence of model of SS onmice’s drinking a mount,thymus,spleen index,TNF-α、IL-6of submandibulargland and IgG of s erum mar kers,we basically studied the mechanism ofZeng Ye Runzao decoction treating SS and applied experimental basis forthe clinical treatment.Methods:Those BALB/c mice which were killed by cervical dislocationwere taken out both sides of submandibular gland,get rid of the capsule andlymph node,weighting,cutting into pieces and putting into homogeratemachine with lysate.Then these tissues were centrifuged in the4℃low-temperatured centrifuge machine,abandoning upper fat layer, keepingsupernatant,getting autoantigen protein which was diluted in to800μg/mlwith sterilization PBS and added freund’s complete adjuvant to be anoil-water mi xed emulsion.70BALB/c mice were randoml y divided into7groups by digital random table:the nor mal group,Freund’s completeadjuvant group,model group,low dose of irradiation of Zeng Ye Runza odecoction,mi ddle dose group,high dose group and total glycosides of peonygroup.The mice from the model group,3Chinese medicine groups a nd totalglycosides of peony group were injected antigen of0.05ml int o theiringuinal of every side on the day before study began,while the mice fr o mFreund’s complete adjuvant group we re also injected the sa me dose ofCFA.On t he14t hday of the st udy, the mice from the model group,3Chinesemedicine groups and total glycosides of peony group wer e injected antigenof0.05ml into their inguinal of every side in order to boostimmuniza tion,while the mice from Freund’s complete adjuvant group werealso injected the sa me dose of CFA. On the2n dand7t hday after theinjection,the mice fr om the Fr eund’s complete adjuvant group,the model group,3Chinese me dicine group and total glycosides of peony group wereinjected diphtheria tetanus and pertussis combined vaccine to ma ke thesubmandi bular gland appeared si milarly as SS di d. By gavage,the3Chinesemedicine groups were given the particle decoction;the nor malgroup,freund’s complete adjuvant group, and model group wer e givendeionizes water;the total glycosides of peony group were given the totalglycosides of peony.During60days the drinking a mount of mice wasmeasured every other day, and after that,those mice were killed to weightthymus and spleen calculating the weight index,measuring the content ofTNF-α、 I L-6in supernatant of submandibular gland and IgG of serummarkers in order to observe the influence of Zeng Ye Runzao decoction indrinking amount,thymus,splee n index,TNF-α、 IL-6of submandibular glandand IgG of serum markers.Results:The Zeng Ye Runzao decoction can increase the drinking amount ofmodel mic e,decrease the thymus, spleen index, TNF-α、 IL-6and IgG.It wasdiscovered from pa thology that the decoction could ease the lymphocyteinfiltration of submandibular gland.Conclusion:The Zeng Ye Runzao decoction can increase the drinkinga mount，decrease the index of TNF-α、IL-6、IgG of model mice by immunityregulation function. With the c ontrol of TNF-α、 I L-6,it decreases indirectlythe proliferation and differentiation of TH subsets,so the reaction ofimmune inflammatory is controlled.Combined with pathologicalsections,the Zeng Ye Runzao decoction can release the lymphocyteinfiltration of submandibular gland of model mice,recover the acinus andcatheters,improve t he function of gland,ease the clinical symptom such asthirsty.This study provides an evidence for the significant function i npathogenesis of SS in TNF-α、 IL-6.As a index of disease activity,thedecoction can also decrease the contents of IgG in the model mice of SS.
