| Objective:We established PCOS rat model to observe the impact of Busehn Huatan Quyv decoction on the estrus cycles,glucose metabolism,fasting serum insulin(FINS),insulin-like grouth factor-1(IGF-1)and the ovarian morphology in polycystic ovary syndrome(PCOS)rat model,the possible mechanism of Bushen Huatan Quyv decoction in the treatment of PCOS were discussed.Methods: 48 SD female rats which were SPF grade 21 days old were selected,After they were fed for 2 days to adapt the environment,when they were 23-day-old the rats were randomly divided into four groups:(1)normal group,(2)model group,(3)metformin group,(4)traditional chinese medicine group,12 per group.the normal group rats were injected subcutaneously 0.2ml soybean oil everyday,while other groups were induced by subcutaneous injection of dehydroepiandrosterone(DHEA)6mg/100 g body weight,mixed with 0.2ml soybean oil for continous 20 d.All rats were given the different drugs for three weeks after the PCOS models were succeed,then based on the result of vagial smear and OGTT test,the blood was obtained and separated from the abdominal aorta after the rats were anesthetized with 10% cloral hydrate(0.4ml/100g)to measure;the ovaries were fixed in neutral buffered formalin(10%)to distinguish the changes of ovarian tissue morphology in light microscopy.Results:(1)The result of vaginal exfoliated cytology: normal group rats' ovulation obstacle was the lowest,showing regular estrus,while model rats' ovulation obstacle was the highest,having irregular estrus,showing long estrous cycle or persistently estrus or diestrus,After lavage treatment,not only traditional chinese medicine group but also metformin group had different degree improvement.(2)The result indicated that there was a downturn about the weight in the treatment groups,but the weight among the rats in different groups was little difference,there was no statistically significant(P>0.05).According to the result,the weight of ovary in traditional chinese medicine group and metformin group were lower than model group which indicated significant difference(P<0.05),but traditional chinese medicine group and metformin group had similar efficancy in ovarian weight,showing no significant difference between the two groups(P>0.05).(3)As well as the traditional chinese medicine group,the the target of FBG and AUC level in the metformin group were decreased significantly,there was a obvious difference compared with the model group(P<0.01),but the two groups showed similar function in the matter of regulating sugar metabolism,there was no striking difference between traditional chinese medicine group and metformin group(P>0.05).(4)compared with the model group,the serum FINS and IGF-1 level of traditional chinese medicine group and metformin group decreased obviously which was a significant difference(P<0.01),traditional chinese medicine group was higher than metformin group about the indicator of FINS and IGF-1,which indicated significant difference(P<0.01 or P<0.05).(5)The changes of ovarian tissue morphology: normal group:There were a certain number of corpus luteum and follicles at different period of development which were clear in cell structure and in alignment.It was obvious that the granule cells form were complete;model group:Most of follicles showed cystic expansion,with few loose granule cells and disappearing corpus luteum;metformin group: Some cystic sinus follicle was in sight,corpus luteum and disorganized granule cell layers were more than model group;traditional chinese medicine group:There were fewer cystic follicles and more corpus luteum.It was obvious that mature follicle or signs of ovulation and granule cells which were morphological integrity and in alignment.Conclusion:(1)Busehn Huatan Quyv decoction can recover estrus cycles and improve ovarian tissue morphology of PCOS rats.(2)Busehn Huatan Quyv decoction can improve glucose metabolism disorder of PCOS rats.(3)Busehn Huatan Quyv decoction can obviously decrease the serum FINS and IGF-1level of PCOS rats. |
