| Objective To study Guiling pa'an pill on PD rat neural stem cell transplantation after the effect of proliferation,migration and the influence of the midbrain black mass DAT,Brn-4.Explore the turtle Guiling pa'an pillimproved PD rat model of neural stem cells in the brain survival possible mechanism;For turtle Guiling pa'an pilltheory of solid scientific basis for treatment of Parkinson's disease,clinical application,achieved a certain economic and social benefits.Method Preparation of pregnant mice,will kill rats at 14 d,aseptic conditions to take out the fetal brain,will the midbrain ventral brain tissue preparation of single cell suspension liquid,inoculated into the culture bottle.Develop the second generation of neural stem cells suspension in climbing 48 forward Br DU positive test results shows that the marking is successful,and set aside.Will build successful PD model rats were randomly divided into blank group,model group,beauty ba group,turtle current Ann pill small dose group,middle dose group and high dose group6.After anesthesia,fixed on the brain stereotactic apparatus,adopting two transplants,located in damaged ipsilateral striatum of rats,steps such as PD rat model is established.Same amount of saline blank group of transplantation,the rest of the group transplanted into the marked success of neural stem cells.Postoperative intraperitoneal injection of 0.5 ml(160 mg/kg)of penicillin,continuous injection of 7 d,single conventional feeding in a cage.From transplantation,at 28 d of groups of animals APO(calculated on to 0.25 mg per kilogram of body weight)rats induced by rotation behavior observation and record in detail.10 min after injection start counting,record the number of rotating circle within 30 minutes.Fluorescence microscope to observe: to observe the behavior changes of rats after rats respectively for each group to be put to death in the brain,the midbrain and striatum coronary frozen section,under the fluorescence microscope to observe the survival of the transplanted cells in rats,and to count on the Brdu positive cells,groups of frozen section after immunohistochemical method to detect DAT,Brn4 expression.Result Compared with the blank group,model group,turtle Guiling pa'an pilllow dose,middle dose and high dose group,the beauty she group rats rotating laps period increased significantly(P?0.05).Compared with model control group,turtle Guiling pa'an pilllow dose,middle dose and high dose group,the beauty she rat midbrain black mass Brdu increased significantly(P?0.05).Compared with model control group,turtle current in Ann bolus dose and high dose group,the beauty she rat midbrain black mass Brn-4 levels increased significantly(P < 0.05).Compared with model control group,turtle current in Ann bolus dose and high dose group,the beauty she rat midbrain black mass DAT content increased significantly(P < 0.05).Conclusion1 Through the 6-OHDA midbrain nigra compacta single-point injection can get ideal animal model of PD.2 Turtle current palmer Ann bolus dose groups after treatment in the rotation period of turns 28 days old rats significantly higher than that of high dose group,low dose group,may be related to turtle current in Ann bolus doses can reach the effective blood drug concentration.Anyhow turtle Guiling pa'an pillcan significantly reduce the Parkinson disease rats by the rotating frequency induced by morphine,o period is associated with turtle current Mr Ann amount of pills.3 According to the neural stem cells to Brdu detection result,turtle Guiling pa'an pilllow dose,middle dose and high dose group,the beauty she can improve the rat midbrain black Brdu content.Current and turtle in Ann bolus dose group,low dose and high dose were superior to beauty ba group,to the best dose.Turtle antelope palmer Ann pill,beauty she may by adjusting the related factors to improve the survival rate neural stem cells.4 Through the Brn-4 groups of rats had,DAT content,can be found that turtle current dose and high dose,beauty in Ann pill ba could improve the rat midbrain black Brn-4,DAT content,say by Parkinson's disease rat midbrain black mass Brn-4,DAT content may be a turtle current Ann pill,beauty she play a role of treatment of one of the mechanisms. |
