Objective: To observe the effect of Guiling pa’an Wan on the expression of apoptosis associated protein that is the content of Bcl-2 and Bax in substantia nigra of Parkinson’s disease rats’ s model,and then to explore and analysis the possible way and target of Guiling pa’an wan in the treatment of Parkinson’s disease from the angle of apoptosis,is to provide the experimental basis for the multi-level and multi-angle mechanism study on the efficacy of Guiling pa’an Wan.Methods:The research group prepared PD rat model which was induced by 6-hydroxydopamine(6-OHDA) with stereotactic technique, and then induced the praxiology change of rats which could judge the model was successful or not with apomorphine(Apomorphine, APO).The successful model rats were randomly divided into 5 groups: Guiling pa’an wan high dose group(high dose group), Guiling pa’an wan middle dose group(middle dose group), Guiling pa’an Wan low dose group(low dose group), Madopar group and Guiling pa’an Wan model group(model group),which was respectively given high dose,middle dose,low dose of Chinese medicine liquid of Guiling pa’an Wan,physic liquor of Madopar and the same volume saline by avage.At the same time, the sham operation group was the control group, which was given normal saline by gavage. The corresponding drug or water was fed for 30 days two times a day. The specimens were observed by TUNEL method the changes of nigral apoptosis, were observed the expression of Bcl-2, Bax protein content in nigral neurons by immunohistochemistry technique and the expression ratio of Bax/Bcl-2.Results: 1.the result of each group SD rats after modeling induced by APO:SD rats model successful rate is 42% in this study. 2. the result of each group SD rats of nigral apoptosis by TUNEL: compared with the model group, the percentage of apoptosis of the high, middle, low dose group and Madopar group was significantly lower than that in the model group(P<0.05), and the model group showed a large number of positive cells;compared with Madopar group, the percentage of apoptosis of the middle, low dose group was significantly higher than that in the Madopar group(P< 0.05), the percentage of apoptosis of the high dose group was slightly higher than that in Madopar group, but there was no statistical difference(P>0.05); the comparation of the three traditional Chinese medicine was statistically significant for the differences(P<0.05), which showed that inhibition on cell apoptosis was more obvious with dosage increase. 3.the result of expression Bcl-2, Bax positive cells and the ratio of Bcl-2/Bax of g each group SD rats of nigral apoptosis: compared with sham operation group, the number of Bcl-2、Bax positive cells of model group rats in the substantia nigra significantly increased(P<0.05), the ratio of Bcl-2/Bax significantly decreased(P<0.05);compared with the model group, the number of nigral Bax positive cells of the high, middle, low dose group and Madopar group significantly decreased(P<0.05), but the number of nigral Bcl-2 positive cells and the ratio of of Bcl-2/Bax increased(P<0.05); compared with Madopar group, the number of Bcl-2 positive cells of high dose group was slightly higher(P>0.05), the number of Bax positive cells of high dose group was obviously lower than that in Madopar group(P<0.05), the ratio of Bcl-2/Bax of high dose group was obviously higher than that in Madopar group(P<0.05); compared with middle dose group and low dose group, the number of nigral Bax positive cells of high dose group decreased, the number of Bcl-2 positive cells and the ratio of Bcl-2/Bax significantly increased, the difference was statistically significant(P<0.05).Conclusion: 1.Guiling pa’an pill can protect the nigral dopamine neuron of PD rats model. 2.Guiling pa’an Wan might suppress the apoptosis of the dopaminergic neuron via improving protein content of Bc1-2 and the ratio of Bc1-2/Bax, but decreasing Bax protein content. 3.Guiling pa’an Wan has a dose effect relationship in the process of inhibition on nigral dopaminergic neuronal apoptosis of PD model rats.
