Objective: To establish a model of hypoxia-induced recurrent upper respiratory tract infection induced by the inactivated vaccine of H5 subtype of cyclophosphamide,and to compare with paracetamol plus calibarlin and modified xiaochaihu decoction.The effect of modified xiaochaihu decoction on the treatment of recurrent upper respiratory tract infection with low immune function is to provide experimental and theoretical basis for the treatment of repeated upper respiratory tract infection with modified xiaochaihu decoction.Methods: 84 healthy Kunming mice were divided into 7 groups,12 rats in each group,and then labeled with each group.The mice were divided into blank group,model group,Xiaochaihu Tang group,western medicine group,modified xiaochaihu decoction low dose group,modified xiaochaihu decoction middle dose group,Modified Xiaochaihu Tang high dose group.First,three days of adaptive feeding,repeated attacks of upper respiratory tract infection modeling,a total of 7 days,using intraperitoneal injection of cyclophosphamide combined with virus intranasal modeling: the model group,Xiaochaihu Tang group,Western medicine group,modified xiaochaihu decoction low,middle and high dose group were treated with cyclophosphamide intraperitoneal injection of cyclophosphamide 0.0016 g / 20(g * d)for 3 days,the sixth day of cyclophosphamide strengthening once,The corresponding group of intraperitoneal injection of the same amount of saline,the seventh day to the virus intranasal,blank group drops the same amount of saline.After the end of modeling,the blank group and the model group were treated with normal saline and the dosage was 10 ml / kg.Xiaochaihu Tang group was treated with Xiaochaihu Tang and 30.42 g crude drug / kg.The western medicine group was treated with acetaminophen and Ribavirin solution,the dose of 0.01 ml / g;modified xiaochaihu decoction low dose group was treated with the low dose of liquid,the dose of 6.63 g crude drug / kg;The middle dose group was treated with the middle dose of liquid,the dose of 19.89 g crude drug / kg;The high group was treated with high dose of liquid,the dose of 33.15 g crude drug / kg.Continuous administration of 7 days,once a day.After the last administration of fasting,can not help but water,drawn.First with throat swabs to take mouse saliva samples,the SIg A content was measured by ELISA;Eyeball method blood,the blood into the centrifuge tube,until the blood coagulation after centrifugation(3000rpm,centrifugal 10min),the serum levels of Ig M and Ig G were measured in the serum of each group;The noses were fixed in 10% formaldehyde solution and embedded in paraffin.The sections were stained with HE and the histopathological images were collected.SPSS19.0 statistical analysis software for data processing.Results: Changes of SIg A in mice:Compared with the blank group,model group,western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly lower,the difference was statistically significant(P<0.05).Compared with the model group,the western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly higher,the difference was statistically significant(P< 0.05).Compared with the Xiaochaihu Tang group and the western medicine group,modified xiaochaihu decoction high dose group was significantly higher then them,the difference were statistically significant(P< 0.05).Mice Ig M content changes:Compared with the blank group,model group,western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly lower,the difference was statistically significant(P< 0.05).Compared with the model group,the western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly higher,the difference was statistically significant(P< 0.05).Compared with the Xiaochaihu Tang group,modified xiaochaihu decoction high dose group was significantly higher,the difference was statistically significant(P<0.05).Compared with the western medicine group,modified xiaochaihu decoction high and middle dose groups were significantly higher,the difference was statistically significant(P< 0.05).Mouse Ig G content changes:Compared with the blank group,model group,western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly lower,the difference was statistically significant(P< 0.05).Compared with the model group,the western medicine group,Xiaochaihu Tang group and modified xiaochaihu decoction groups were significantly higher,the difference was statistically significant(P< 0.05).Compared with the Xiaochaihu Tang group,modified xiaochaihu decoction high dose group was significantly higher,the difference was statistically significant(P<0.05).Compared with the western medicine group,modified xiaochaihu decoction high,middle and low dose groups were significantly higher,the difference was statistically significant(P< 0.05).Pathological changes: light microscopic observation of each group of tissue sections showed: blank group mice nasal mucosa structure is normal,no inflammatory changes.The mice in the model group showed significant swelling of the nasal mucosa,mucosal congestion and edema.The nasopharyngeal mucosa of the drug group was improved in different degree,but the pathological changes of the modified xiaochaihu decoction group were better than those of the other two groups.Conclusion: 1.modified xiaochaihu decoction is effective in the treatment of cyclosporine-induced recurrent upper respiratory tract infection in mice induced by trivalent inactivated vaccine against H5 subtype of cyclophosphamide.2.modified xiaochaihu decoction treatment of repeated upper respiratory tract infection may be through the regulation of SIg A,Ig M and Ig G immune function to achieve the state.
