Purpose: The study was designed to observe the effect of reforcing the kidney and spleen,detoxification and leaching pharyngeal method on TH1/TH2 in IgA nephropathy rats,and further to explore the mechanism of this method in the treatment of IgA nephropathy in order to provide experimental evidence for the clinical treatment of IgA nephropathy.Methods: Male and healthy eighty Wistar rats(180 ± 20g)were adapted to the environment for a week,then randomly hierarchical divided into blank group(n = 10)and model group(n = 70).The rats in the blank group were given with saline by using gavage,subcutaneous injection of saline and tail vein injection of saline,the rats in the model group were treated with Bevine serum albumin(BSA)by using gavage,subcutaneous injection of carbon tetrachloride(CCL4),and tail vein injection of lipopolysaccharide(LPS)to prepare IgA nephropathy model rats.IgA nephropathy model was successfully divided into model group,Lotensin control group,reinforcing kidney and spleen,detoxification and leaching pharyngeal method medicine high,medium and low dose groups.The rats in Lotensin control group were given with 6mg/(Kg·d)lotensin by using gavage.The rats in the low dose group of detoxification and leaching pharyngeal medicine were treated with 4ml/(kg ·d)(equivalent to 8g/kg of traditional Chinese drink tablets)by using gavage,the rats in the medium group were treated with 4ml/(kg·d)(equivalent to 16g/kg of traditional Chinese medicine drink tablets),the rats in high-dose group of Chinese medicine decoction were treated with4ml/(kg·d)(equivalent to traditional Chinese medicine tablets 32g/kg)by using gavage,the rats in blank group and model groups were given with daily saline 4ml/each one.The experiment was for 8 weeks.The indices detection:(1)the general statement of the rats.(2)24h urine erythrocyte count and 24 h urine proteinuria detection of the rats.(3)Serum renal function and liver function test of the rats.(4)Detection of TH1/TH2 in renal tissue of the rats.(5)Detection of the positioning expression of TH1/TH2 in renal tissue of the rats.(6)Detection of the deposition of IgA complex in renal tissue of the rats.Results:(1)Twenty-four rats died during the experiment.The rats were included that twelve rats were sacrificed in order to verify the model.Ten rats died during the modeling process and two rats died during the course of treatment in the lotensin control group.(2)The rats in the blank group had been in good condition and no obvious abnormality during the whole experiment.Rats in each model group were treated with subcutaneous injection of carbon tetrachloride showing that the spirit of malaise,hair clutter without glossy,mouth and nose secretions,diarrhea,weight loss and other performance.After 2 weeks of treatment,the rats in the treatment group were improved,decreased diarrhea symptoms and gained weight.(3)There were no hematuria and proteinuria symptoms in rats of the blank group.The 24 h urine erythrocyte count and 24 h urine protein were significantly increased in rats of the model group,after the rats was treated with reinforcing kidney and spleen,detoxification and leaching pharyngeal method,The 24 h urine erythrocyte count and 24 h urine protein were significantly reduced(p<0.05).Chinese medicine group is better than lotensin control group.(p<0.05)(4)Compared with rats in the blank group,the level of IFN-? in renal tissue of IgA nephropathy rats was significantly lower and the level of IL-4 was significantly increased(p< 0.05).(5)The level of IFN-? was significantly up-regulated(p < 0.05)and the level of IL-4 was significantly decreased in the treatment group(p<0.05),Chinese medicine group is better than lotensin control group.(p<0.05)(6)There was no significant difference in renal function and liver function among the all experimental groups(p>0.05).Conclusions:(1)TH1/TH2 balance in renal tissue of IgA nephropathy model rats drifts to TH2.(2)The level of TH1 was significantly increased after treatment,indicating that TH1 was positively correlated with the condition;TH2 was down-regulated after treatment,indicating that TH2 was negatively correlated with the condition,the results demonstrated that TH1/TH2 abnormal expression was involved into the incidence of IgA nephropathy,leading to the occurrence of IgA nephropathy.(3)Comparison of efficacy of the all treatment groups,the possible mechanism of reinforcing kidney and spleen,detoxification and leaching pharyngeal medicine underlying delay the development of IgAN was by regulating the balance of THI/TH2 in renal tissue of IgAN model rats. |
