[Objective]To investigate the regulation of "Qi Yao Ji Jin Gruel" medicated diet on intestinal tract immune dysfunction of rats with 5-fluorouracil induced.gastrointestinal mucositis.Understanding the effect of preventing and curing of 5-fluorouracil induced gastrointestinal mucositis by medicined diet,to probe a new way to prevent and cure the alimentary tract mucositis after chemotherapy.[Methods]Used a single dose of 5-fluorouracil at 150mg/kg to cause reproducible gastrointestinal mucositis.48 healthy Wistar rats were randomly divided into three groups by weight:model group;medicined diet group;normal control group.Monitor the dynamic change of general condition?body weight?food intake and diarrhea in rats during the experiment.Rats were killed at the 1st and 7th days after chemotherapy with 10 each for each group,to detect:(1)the level of SlgA(2)the changes of CD3+?CD4+?CD8+T lymphocyte cells and the ratio of CD4+/CD8+ of rat intestinal.(3)intestinal mucosa Pathomorphology observation.(4)gastric mucosal Pathomorphology observation.All the experimental datas were analyzed by SPSS 16.0.[Results]1.Scores of general condition in ratsThe scores of general condition in rats of model group and medicined diet group was significantly higher after chemotherapy than before,and it is also higher than normal control group at the same period.Since the second day after chemotherapy,the scores of general condition in rats of model group was significantly higher than the medicined diet group(P<0.05 or P<0.01),and the trend continued until the end of the experiment.2.The changes of body weight,food intake,diarrhea in rats2.A Body weight:The rats of normal control group showed a progressive increase in body weight after chemotherapy.The body weight of rats in model group and medicined diet group were decreased.The body weight in the rats of medicined diet group recoverd to normal levels from the 4 days after chemotherapy.The body weight in the rats of model group was lower than medicined diet group from the 2 days after chemotherapy until the end of the experiment.2.2 Quantity of food intakeThe quantity of food intake in rats of model group and medicined diet group decreased significantly from 1-3days after chemotherapy and it is increased from the 4 days until the endof the experiment,2.3 Diarrhea rates:the rats of model group and medcined diet group showed diarrhea after 24 hours of 5-Fu injection and diarrhea reach to peak at 72 hours after chemotherapy.3.The intestinal tract immune dysfunction3.1 T lymphocyte cells and subgroups:At the 1st days after chemotherapy,the expression of T lymphocyte cells and subgroups and CD4+/CD8+ ratio in medicated diet group and model group were significant differences compared with normal group(P<0.05 OR P<0.01);However,there were notably increased of the expression of CD4+T lymphocyte cells and CD4+/CD8H+ratio in medicated diet group than that in model grorup(P<0.05 OR P<0.01).At the 7th days after chemotherapy,no obvious difference of the expression of CD3+,CD4+ was found between medicated diet and normal group(P>0.05).But the expression of CD3+,CD4+,CD8+T lymphocyte cells and CD4+/CD8+ ratio in medicated diet had significant differences than that of model group(P<0.05 OR P<0.01).3.2 The level of SlgA:Compared with the control group,the lever of SlgA of the medicined diet and model group was apparently decreaded at lday after chernotherapy;At 7day after chemotherapy,the lever of slgA of medicined diet was decreased compared with normal group but much higher than that of model group,4.Intestinal mucosa pathomorphology observationcompared with the normal group,different pathomorphological changed in intestinal mucosa of medicined diet and model group after chemotherapy.Intestinal mucosa obvious atrophy,the epithelial cells of mucosa necrosis,off,goblet cells increased,heavy inflammation,submucosa thinning and loosen,muscularis thinning or fracture.5.gastric mucosal Pathomorphology observationThe gastric mucosa of rats in nomal control group was nomal under Light microscope,the surface of gastric tissue was smooth and the texture was clear and there was no inflammatory reaction and damage.There was significant damage of the gastric mucosa in rats of model group.It showed dark red congestion,edema and bleeding points was scattered,The mucosa cells has a significantly degeneration?necrosis or loss and there was inflammatory response in lamina propria.There was a small amount of bleeding and erosion in gastric mucosa of medcined diet group,and there was a small amount of inflammatory exudate in the surface of gastric mucosa under Microscope,and there was degeneration and necrosis at the superficial mucosal cells.[Conclusions]It is suggested that medicated diet not only could prevent and cure the alimentary tract mucositis after chemotherapy,also could accelerate the hyperplasy of lymphocyte cells in the intestinal tract,prevent the decrease of the lever of SlgA,improve the proportion of T-lymphocyte subsets,and eventually improve the immunity function of intestinal tract of rats with chemotherapy-induced gastrointestinal mucositis.
