Objective: Based on the effectiveness of acupuncture in treating post traumatic stress disorder(PTSD),this research tested and analysed the changes of behavior,learning and memory function and electrical activity of the brain of PTSD model rats;and further investigated the effects of "Dispersing Liver and Regulating Spirit" Acupuncture Method on PTSD rats' neurological function,and provided the mechanism basis for research and clinical applications of acupuncture intervening PTSD.Method: 60 healthy male SD rats of 6 weeks old were screened out by Morris water maze experiment and then randomly divided into blank control group,model group,grasp group,acupuncture group and paroxetine hydrochloride group after one week adaptive feeding.Apart from the blank control group,all the rats with PTSD were created model by "electric shock + incarceration" for 7 days.Paroxetine hydrochloride group giving paroxetine hydrochloride solution Po(10ml·kg·day)intragastrically treatment 1 hour before modeling;acupuncture group treated by "Dispersing Liver and Regulating Spirit" acupuncture method on the points “baihui”,“neiguan”,“shenmen”,“taichong” 1 hour before modeling;grasp group was grasped as the same way as other intervention groups.After modeling,all intervention groups continued to be treated for another 5 days.After that,all groups were tested for their behavior,learning and memory,eeg.Results:1 Changes of general signs in rats:(1)Weight: all rats' weight showed a rise tendency as a whole,the weight of the blank control group increased fast and stable,and in good condition;model group and grasp group from the third day of modeling the weight increased slowly;rats in the treatment groups increased slowly in weight during modeling,but after that increased quickly,and showed well nutritional status.(2)Other changes: the blank control group showed bright wool color,normal spirit and activities;as decreased food intake,model group were emaciated,no gloss in wool,spirit languish;grasp group was similar with model group;rats in the treatment groups were recoveried compared with model group,but paroxetine hydrochloride group appearedsleepiness in the late treatment.2 Behaviour changes of rats in each group(1)Comparison of open-field testCompared with blank control group,the model group showed significantly reduce in Horizontal crossing number(P <0.01),significantly reduce in vertical times(P <0.01),apparently increase in dejection amounts(P <0.01);there was no significant difference between the grasp group and model group(P >0.05);compared with the model group,each treatment group were significantly increased in total distance(P <0.01,P <0.01),significantly increased in vertical times(P <0.01,P <0.01),and apparently decreased in dejection amounts(P <0.01,P <0.01);there was no significant difference between the treatment groups(P >0.05).(2)Comparison of Morris water maze(1)Place navigation test: the escape latency of each group was reduced in different degrees.Compared with the blank control group,the average escape latency time in model group was significantly prolonged(P <0.05),especially in third and forth days(P <0.01,P<0.01);there was not statistically significant(P >0.05)between grasp group and model group;compared with grasp group,the average escape latency time in acupuncture group and paroxetine hydrochloride group were shortened(P <0.05,P <0.05),paroxetine hydrochloride group showed significantly shortened(P <0.01)since the fourth day,but acupuncture group was significantly shortened(P <0.01)since second day.(2)Space exploration test: Compared with the blank control group,the average escape latency time in model group was significantly prolonged(P <0.01),across platform times were significantly reduced(P <0.01),through the effective area times were significantly reduced(P <0.01);there was not statistically significant(P >0.05)between grasp group and model group;compared with grasp group,compared with the grasp group,the average escape latency time in each treatment group was significantly shortened(P <0.01,P <0.01),across platform times were significantly increased(P <0.01,P <0.01),the times of through the effective area were significantly increased(P <0.01,P <0.01);there was no significant difference between the treatment groups(P >0.05).3 Comparison of new object recognition testCompared with the blank control group,the model group showed lower DI%(P <0.05);there was no significant difference between grasp group and model group(P >0.05);compared with the grasp group,each treatment group had increased DI%(P <0.05,P <0.05);there was no significant difference between the treatment groups(P >0.05).4 Changes of EEG in each group(1)Comparison of Alpha waveCompared with the blank control group,? power value of model group was significantly decreased(P <0.01)in bilateral cerebral hemisphere;there was no significant difference between grasp group and model group(P >0.05);compared with the grasp group,? power value was significantly increased(P <0.01,P <0.01)in each treatment group;there was no significant difference between the treatment groups(P >0.05).(2)Comparison of Beta waveCompared with the blank control group,? power value of model group was significantly increased(P <0.01)in bilateral cerebral hemisphere;there was no significant difference between grasp group and model group(P >0.05);compared with the grasp group,? power value was significantly decreased(P <0.01,P <0.01).in each treatment group;there was no significant difference between the treatment groups(P >0.05).(3)Comparison of Delta waveCompared with the blank control group,? power value of model group was significantly increased(P <0.01)in bilateral cerebral hemisphere;there was no significant difference between grasp group and model group(P >0.05);compared with the grasp group,? power value was significantly decreased(P <0.01,P <0.01).in each treatment group;there was no significant difference between the treatment groups(P >0.05).(4)Comparison of Theta waveCompared with the blank control group,? power value of model group was significantly increased(P <0.01)in bilateral cerebral hemisphere;there was no significant difference between grasp group and model group(P >0.05);compared with the grasp group,? power value was significantly decreased(P <0.01,P <0.01).in each treatment group;there was no significant difference between the treatment groups(P >0.05).Conclusion:(1)Rats with PTSD had abnormal neurobehavioral changes and impaired learning and memory function.(2)Rats with PTSD had abnormal ?,?,?,? changes in bilateral cerebral hemispheres,indicating that brain neuronal activity and energy metabolism were disordered,cerebral cortexmay exist dysfunction(such as memory,emotion,behavior etc.)and abnormal changes,and the abnormal electrophysiological damage is likely to be one of the important central mechanism of PTSD.(3)"Dispersing Liver and Regulating Spirit" acupuncture method can significantly improve the abnormal EEG power spectrum value,abnormal behavior and impaired learning and memory function of rats with PTSD;acupuncture group rats were different from model group rats in brain waves,that may be important mechanisms of " Dispersing Liver and Regulating Spirit" acupuncture method therapy for PTSD.
