| Rhamnella gilgitica is the dried xylem of the stem of the Rhamnella gilgitica Mansf.et Melch.With cooling blood and detumescence,anti-inflammatory effect,people used to treat rheumatoid arthritis,grasserie,alpine blood diseases and other diseases,and have the advantages of good curative effect,less adverse reactions,long-term use etc.In recent years,a large number of clinical studies have reported that there are significant anti-inflammatory and analgesic effects,but also have a good therapeutic effect on osteoarthritis.In this paper,we tried to explore the analgesic and anti-inflammatory effects of the extracts from the different parts of Rhamnella gilgitica.1、Analgesic and anti-inflammatory effects of different extracted parts of Rhamnella gilgiticaSelecting the Rhamnella gilgitica as the research object,firstly,the extraction by refluxing method,then the concentration of extracts obtained extract.Secondly,n-butanol and ethyl acetate were selected for the extraction,and the three kinds of extracts were extracted from n-butanol,ethyl acetate and the remainder of the other two groups.The antioxidant capacity of different extracts were determined by Prussian blue method,Fenton method and DPPH method,selected mice ear swelling model to examine the inhibitory effects of different extracts on acute inflammation,to observe the analgesic effect of the Rhamnella gilgitica of different extracts by acetic acid in mice model.The results show that Matsuike butanol and ethyl acetate extracts has certain ability to scavenge free radicals,and the scavenging rate and sample concentration had obvious dose effect relationship,so that different extracted parts of Rhamnella gilgitica has certain antioxidant capacity;pharmacological experiment showed that matsuike n-butanol and ethyl acetate extracts to after the drug could inhibit xylene induced mouse ear swelling degree,and reduce the writhing times of mice(P<0.05 or 0.01),extraction of n-butanol,ethyl acetate extract has certain analgesic and anti-inflammatory effects.2、Study on Optimization of extraction technology of total flavonoids from Rhamnella gilgitica and its effect on rat model of osteoarthritisUsing ethanol reflux extraction of total flavonoids from Rhamnella gilgitica in extraction,solid-liquid ratio,extraction time,ethanol concentration,the extraction rate of the number of total flavonoids were investigated by single factor experiments,identified 4 levels of orthogonal experiment,the optimal extraction process of total flavonoids in the extract of 60% ethanol solution,solid-liquid ratio 1:30,extracting 2 times,extracting time 3 h,through the verification of the process shows that the process has good reproducibility,high extraction rate.Then the model of osteoarthritis in rats,the content of 1L-1 by NO level,joint effusion and TNF-determination of alpha beta after administration of animal serum,and the pathological section method to investigate medicine treatment effect of total flavonoids of Rhamnella gilgitica and so on osteoarthritis.The results showed that each group of IL-l rats joint effusion,serum alpha beta,TNF-content level was lower than that of model group(P<0.05 or 0.01),and the pathological sections showed that the cartilage surface is smooth,cartilage cells were arranged in order.The results showed that total flavonoids of Rhamnella gilgitica have a certain therapeutic effect on osteoarthritis.This paper on the different extracts were prepared,and investigated its in vitro antioxidant activity and the anti-inflammatory and analgesic effect of total flavonoids of Rhamnella gilgitica,then were extracted,the extraction process was optimized,the rat osteoarthritis model to study the therapeutic effect of inflammation.It was found that the extract of Rhamnella gilgitica has certain anti-inflammatory,analgesic and antioxidant effects,and its total flavonoids have a certain inhibitory effect on osteoarthritis.This study provides a theoretical basis for the clinical use of Rhamnella gilgitica in the treatment of inflammatory diseases and relief of pain symptoms. |
