Objective Total flavonoids of Chuzhou chrysanthemum(TFCC) is an active partextracted from chrysanthemum dired capitulum.The major effective components inTFCC are diosmetin, apigenin, luteolin, quercetin and acacetin etc. Analgesic andanti-inflammatory experiment of TFCC has not been reported. In this paper, we use avariety of classic anti-inflammatory analgesic model to study the TFCC analgesicanti-inflammatory effects. Through the measurement of NO and PGE2content., andstudy its mechanism.Methods and Results1. TFCC analgesia effectAnalgesic experiments using mice writhing, warm bath, formalin, hot plate method,intracerebroventricular injection (icv); using analgesic morphine and aspirin do positivecontrol experiment to observe the reaction to pain inhibition TFCC.Results can be seen,ig TFCC100,200mg kg-1can significantly reduce thenumber of writhing, formalin-induced pain in mice reduced the role of the first phaseⅡ pain response scores, extending small mouse hot plate licking response latency andhot tails latency that TFCC have significant analgesic effect.Icv TFCC5and10mg kg-1can inhibit writhing inhibition rate can reach52%.The results suggest that, TFCC analgesic effect may have some central mechanisms involved.2. TFCC anti-inflammatory effectExperimental anti-inflammatory ear edema in mice,rat granuloma.Anti-inflammatory drugs naproxen using a positive control, the inflammatory responseobserved inhibition of TFCC.Results can be seen: ig TFCC400mg kg-1, xylene-induced ear edema in micesignificantly inhibited,the inhibition rate can reach36.4%.TFCC at the dose of320mgkg-1, reversed the cotton-induced proliferation of granuloma.The results evidencethat, TFCC has significant anti-inflammatory effect.3. TFCC analgesic effect relationship between NO and PGE2Analgesic experiments, mice eyeball removal of blood, serum and brain tissuelevels of NO and PGE2were used Griess method and UV spectrophotometry. Theresults showed,ig TFCC100,200mg kg-1can obviously increase the content of NOin serum and brain tissue of mice, but the content of PGE2was decreased. The ability ofTFCC to promote the release of NO and inhibit of PGE2in serum and brain tissue.Concluded TFCC has significant analgesic effect which may be related to increasingthe serum and brain tissue NO release, reducing the generation of PGE2on,the centralmechanism is at least partially involved in the analgesic effect of TFCC. TFCC hasobvious anti-inflammatory effect.
