Objective The experiment focuses on the study of extraction and purification of total flavonoids from Polygonum amplexicaul e var.sinense. At the same time it studies the effect of anti-inflammation and analgesia of total flavonoids by the optimal extraction and purification.Methods The experiment compares the process of traditional extraction and microwave extraction and with the extraction rate of total flavonoids as the index it determines the content of total flavonoids by using ultraviolet and visible spectrophotometer. Using the single factor experiment and the orthogonal design experiment, the best method of extraction of total flavonoids from Polygonum amplexicaule var.sinense is confirmed. With the help of static and dynamic adsorption experiment, it can investigates the optimal purifying technology of total flavonoids with the macroporous resin. Xyl induces the swelling of ears of the mice. This can be used to test the effect of anti-inflammation of the total flavonoids of Polygonum amplexicaule var.sinense. With the help of hot-plate and writhing-test, the analgesic effect can be tested.Results The experiment compares the two methods, traditional extraction andmicrowave extraction, under the optimal condition. It’s found that the each extraction rate is 2.81% and 3.27%. In the aspect of extraction rate, microwave extraction is higher than traditional extraction, microwave extraction is effectiver than traditional extraction too. Macroporous-adsorption-resin purifies the total flavonoids of Polygonum amplexicaule var.sinense in the experiment. So the best method to the purifying of the total flavonoids is: the concentration of the total flavonoids solution 1.826 mg/m L, the speed sampling of the sample 2m L/min, put the solution on the adsorption-resin. The eluting solution is ethanol(60%), 2m L/min speed for elution. The purity of the total flavonoids of Polygonum amplexicaule var.sinense after purifying is 78.22%, which has obvious effect to inhibite the swelling of mice’s ears. That shows its effect of anti-inflammatory. In the aspect of analgesia, it reduce obviously the writhing of mice and the pain-threshold increases. It confirms its effect of analgesia.Conclusions The method of microwave extraction is effectiver, convenienter and feasibaler than the traditional extraction. With macroporous-adsorption-resin the total flavonoids of Polygonum amplexicaule var.sinense can be purified and after purifying it has some effects on anti-inflammatory and analgesic.
